摘  要:目的 分析细胞质聚腺苷酸化元件结合蛋白3(Cytoplasmic Polyadenylation Element Binding Protein 3,CPEB3)在5种皮肤黑素瘤细胞中的表达情况,并通过下调其在细胞中的表达,评价CPEB3基因对细胞增殖、凋亡、迁移和侵袭能力的影响。方法 利用实时荧光定量PCR(RT-qPCR)和Western blot检测5种黑素瘤细胞株(WM3211、SK-MEL-28、A2058、A375和A875)中CPEB3在mRNA和蛋白水平的表达情况;应用shRNA干扰技术下调细胞中CPEB3的表达,采用CCK-8法、平板克隆形成试验、流式细胞术、划痕法、Transwell迁移和侵袭试验等方法检测CPEB3表达下调对黑素瘤细胞增殖、凋亡、迁移和侵袭能力的影响。结果 CPEB3在5种黑素瘤细胞株中的表达水平从高到低依次为:WM3211、SK-MEL-28、A2058、A375和A875;下调SK-MEL-28细胞中CPEB3的表达后,细胞增殖能力和克隆形成能力显著增强,凋亡率显著降低,细胞迁移和侵袭能力显著提高(均P<0.05)。结论 下调黑素瘤细胞中CPEB3的表达可促进细胞增殖,抑制细胞凋亡,并增强细胞迁移和侵袭能力,提示CPEB3基因可能在黑素瘤恶性进展过程中发挥重要作用。Objective To analyze the expression of Cytoplasmic Polyadenylation Element Binding Protein 3(CPEB3)in five cell lines of cutaneous melanoma,and to evaluate the effect of CPEB3on cell proliferation,apoptosis,migration,and invasion by down-regulating the expression of CPEB3in cells.Methods Real-time fluorescence quantitative PCR(RT-qPCR)and Western blot techniques were used to detect the expression of CPEB3at mRNA and protein levels in five mel-anoma cell lines(WM3211,SK-MEL-28,A2058,A375,and A875).Using shRNA interference technology to down-regu-late the expression of CPEB3in cells,we detected the effects of down-regulation of CPEB3expression on the proliferation,apoptosis,migration,and invasion ability of melanoma cells by CCK-8,plate clone formation assay,flow cytometry,scratch assay,Transwell migration,and invasion assay.Results The expression of CPEB3decreased sequentially in five melano-ma cell lines:WM3211,SK-MEL-28,A2058,A375,and A875cell lines.After down-regulation of the expression of CPEB3 in SK-MEL-28cell,cell proliferation and clonal formation ability were significantly enhanced,while the apoptosis rate was significantly reduced,and the cell migration and invasion ability were significantly improved(P<0.05).Conclusion Down-regulation of CPEB3expression in melanoma cells can promote cell proliferation,inhibit cell apoptosis,and enhance cell migration and invasion.Those results suggest that CPEB3gene may play an important role in the malignant progres-sion of melanoma.
