摘 要:目的优选甘木通强心苷的提取工艺,测定甘木通中强心苷的含量,研究其抗心力衰竭的作用。方法以强心苷含量以及干膏得率为评价指标,采用正交试验优选甘木通强心苷最佳提取工艺。采用紫外光度法测定不同产地不同部位甘木通的强心苷含量。采用冠状动脉前降支结扎法制备大鼠心力衰竭模型,记录各组大鼠心脏血流动力学指标,测定大鼠血清中醛固酮(ALD)、血管紧张素Ⅱ(AngⅡ)、N-前端心钠肽(ANP)和N-前端脑钠肽(BNP)的含量,计算心脏指数。结果甘木通强心苷的最佳提取工艺为料液比为1∶20(质量容积比),乙醇浓度85%,提取温度80℃,超声提取30 min。甘木通根、茎、叶部位的强心苷含量分别为0.25~0.43、0.67~0.86、0.41~0.67 mg·g^(-1)。与假手术组比较,模型组左室收缩压(LVSP)、左室内压最大上升速率(+dp/dt_(max))、左室内压最大下降速率(-dp/dt_(max))和心率(HR)显著下降(P<0.05),左室舒张末压(LVEDP)显著升高(P<0.05),大鼠血清中ALD、AngⅡ、ANP和BNP含量明显上升(P<0.05),全心质量指数(HMI)和左心室质量指数(LVMI)明显升高(P<0.05)。与模型组比较,甘木通高、低剂量组和阳性药卡托普利组大鼠LVSP、+dp/dt_(max)、-dp/dt_(max)和HR显著升高(P<0.05),LVEDP则显著降低(P<0.05);血清中ALD、AngⅡ、ANP和BNP含量显著降低(P<0.05),HMI和LVMI明显降低(P<0.05)。结论所优化的工艺适用于甘木通中强心苷的提取,甘木通中强心苷的含量与药材的部位,以及采集的地区、时间有关;甘木通强心苷对慢性心力衰竭大鼠具有保护作用。Objective To optimize the extraction process of cardiac glycosides from Clematis filamentosa Dunn,and to determine the content and anti-chronic heart failure of cardiac glycosides.Methods Orthogonal test was performed to optimize the extraction process by using the content of cardiac glycosides and the yield of dry extract as indexes.The content of cardiac glycoside in different parts of Clematis filamentosa Dunn from different habitats were detected by ultraviolet spectrophotometry.The chronic heart failure model was established by ligating left anterior descending coronary artery,hemodynamic parameters of rats in each group were recorded.The contents of aldosterone(ALD),angiotensin Ⅱ(AngⅡ),N-terminal atrial natriuretic peptide(ANP)and N-terminal brain natriuretic peptide(BNP)in serum of rats were determined,while cardiac index was measured.Results The optimum extraction process of cardiac glycosides from Clematis filamentosa Dunn were as follows:liquid to material ratio of 1∶20(mass/volume ratio),ethanol concentration of 85%,extraction temperature of 80℃,ultrasound treatment time of 30 minutes.The contents of cardiac glycosides in root,stem and leaves from Clematis filamentosa Dunn were in the range of 0.25-0.43,0.67-0.86,0.41-0.67 mg·g^(-1),respectively.Compared with sham operation group,the left ventricular systolic pressure(LVSP),maximal rates of rise in left ventricular pressure(+dp/dt_(max)),maximal rates of fall in left ventricular pressure(-dp/dt_(max))and heart rate(HR)of the rats in model group were significantly decreased(P<0.05),and the left ventricular end-diastolic pressure(LVEDP)was significantly increased(P<0.05).The levels of ALD,AngⅡ,ANP and BNP in serum were significantly increased(P<0.05),and heart mass index(HMI)and left ventricular mass index(LVMI)were significantly increased(P<0.05).Compared with model group,the LVSP,+dp/dt_(max),-dp/dt_(max)and HR of the rats in high-and low-dose of Clematis filamentosa Dunn groups and positive drug(captopril)control group were significantly in
