摘 要:目的探究葛根素水凝胶对糖尿病大鼠皮肤创面血管形成及愈合的影响。方法用随机数表法将36只糖尿病背部创面大鼠分为对照组、1%葛根素组、2%葛根素组和4%葛根素组,每组9只,分别用磷酸盐缓冲液和不同浓度的葛根素水凝胶涂抹创面,每天1次,连续14 d。于第3、7和14天观察创面愈合情况,完整切取创面组织,HE及Masson染色观察创面组织学变化,免疫组织化学法进行血管内皮细胞生长因子(VEGF)和CD31染色,显微镜下拍照后利用Image pro plus 6.0软件计算积分光密度(IOD)及微血管密度(MVD),Western Blot法检测创面组织VEGF蛋白表达情况。结果与对照组比较,各葛根素组创面愈合情况均较好,以4%葛根素组最明显(P<0.01)。HE及Masson染色结果显示,与对照组比较,各葛根素组在第7、14天时胶原纤维增生明显(P<0.01),并有大量肉芽组织形成。免疫组织化学染色结果显示,与对照组比较,各葛根素组在第7、14天时创面组织内VEGF IOD和MVD均明显增加,具体表现为随时间推移和葛根素浓度增加而显示出更高的VEGF表达和更多的微血管生成(P<0.05)。Western Blot结果显示,与对照组比较,葛根素水凝胶可呈剂量依赖地促进VEGF蛋白的生成(P<0.01)。结论葛根素水凝胶可能以浓度依赖的方式增加创面组织内VEGF及CD31的表达和微血管生成,促进创面组织胶原纤维的沉积,加速创面愈合。Objective To explore the effect of puerarin hydrogel on wound angiogenesis and healing in diabetic rats.Methods Thirty-six diabetic rats with back wound were divided into the control group,1% puerarin group,2% puerarin group and 4% puerarin group by a random number table,with 9 rats in each group.The back wound in diabetic rats was treated with phosphate buffered saline and different concentrations of puerarin hydrogel respectively,once a day for 14 days.The wound healing was observed on 3,7 and 14 days,and wound tissue was removed completely,the histological changes were observed by HE and Masson staining,immunohistochemistry for VEGF and CD31 staining was performed,and the integrated optical density(IOD)and microvascular density(MVD)were calculated by Image pro plus 6.0 software after microscopic photography was taken.The expression of VEGF protein in wound tissue was detected by Western Blot.Results Compared with the control group,the wound healing was better in all puerarin groups,especially the 4% puerarin group(P<0.01).HE and Masson staining results showed that significant proliferation of collagen fibers at 7 and 14 days in puerarin groups was observed(P<0.01),accompanied by the formation of abundant granulation tissue when compared with the control group.Immunohistochemical staining results showed that compared with the control group,VEGF IOD and MVD in wound tissue of each puerarin group were significantly increased at 7 and 14 days,which was specifically manifested as increased VEGF expression and microangiogenesis with the raise of time and the puerarin concentration(P<0.05).Western Blot results show that the puerarin hydrogel could promote the generation of VEGF protein in a dosedependent manner when compared with the control group(P<0.01).Conclusion Puerarin hydrogels may concentration-dependent increase the expressions of VEGF and CD31 and microangiogenesis in wound tissue,and promote deposition of collagen fibers in wound tissue and accelerate wound healing.
