摘 要:目的探讨抗纤益心方(升陷汤化裁)对大鼠心肌细胞肥大模型线粒体质量控制的影响。方法用改良Langendorff灌流法隔离成年大鼠心肌细胞。实验分为正常组、模型组、中药抗纤益心方组、西药辅酶Q_(10)组。用去甲肾上腺素(NE)诱导大鼠心肌细胞肥大模型,ImageJ图像专业分析软件测定心肌细胞表面积,荧光素酶检测心肌细胞ATP表达水平,Real-Time PCR及Western Blot法检测SIRT3、PGC-1α、Mff、Drp1、Pink1、Parkin mRNA及蛋白表达水平。结果与模型组比较,成年大鼠心肌细胞经过NE处理后,中药抗纤益心方干预后大鼠心肌细胞表面积减小,BNP mRNA表达降低,差异有统计学意义(P<0.05);中药抗纤益心方组与西药辅酶Q_(10)组大鼠心肌细胞ATP含量有所增加,差异有统计学意义(P<0.05);中药抗纤益心方组成年大鼠心肌细胞中SIRT3、PGC-1α、Mff、Pink1、Parkin蛋白表达有所升高,差异有统计学意义(P<0.05);Drp1蛋白表达无明显变化,差异无统计学意义(P>0.05);中药抗纤益心方组成年大鼠心肌细胞中SIRT3、PGC-1α、Mff、Pink1、Parkin mRNA表达有所升高(P<0.05),Drp1 mRNA表达有所升高但差异无统计学意义(P>0.05)。结论抗纤益心方能够对线粒体质量控制环节多种蛋白表达进行调控,发挥对扩张型心肌病的保护作用,其机制可能与SIRT3密切相关。Objective To investigate the effect of Kangxian Yixin Decoction(KYD,modified Shengxian Decoction)on mitochondrial quality control in rat models of cardiomyocyte hypertrophy.Methods Adult rat cardiomyocytes were isolated by the improved Langendorff perfusion method.The cells were divided into control group,model group,KYD group and coenzyme Q_(10)group.The cardiomyocyte hypertrophy model was induced by norepinephrine(NE).The surface area of cardiomyocytes was measured by Image J,and ATP expression in myocardial cells was determined by luciferase assay,the mRNA and protein expression levels of SIRT3,PGC-1α,Mff,Drp1,Pink1 and Parkin were detected by Real-Time PCR and Western Blot.Results Compared with the model group,after adult rat cardiomyocytes were treated by NE,the surface area of cardiomyocytes and the expression of BNP mRNA in the KYD group were decreased,and the differences were statistically significant(P<0.05).The ATP content in cardiomyocytes of rats in the KYD group and coenzyme Q_(10)group was increased,and the difference was statistically significant(P<0.05).The protein expressions of SIRT3,PGC-1α,Mff,Pink1 and Parkin in rat cardiomyocytes treated with KYD were significantly increased(P<0.05),while there was no significant change in the protein expression of Drp1(P>0.05).The mRNA expressions of SIRT3,PGC-1α、Mff,Pink1 and Parkin in adult rat cardiomyocytes treated with KYD were increased(P<0.05),while the mRNA expression of Drp1 was increased,and the difference was not statistically significant(P>0.05).Conclusion Kangxian Yixin Decoction can regulate the expression of multiple proteins in mitochondrial quality control and play a protective role in dilated cardiomyopathy.Its mechanism may be closely related to SIRT3.
