摘  要:为了开发一种新型生物活性伤口敷料,以丝素蛋白(SF)和酪胺改性明胶(TG)为基体材料,通过辣根过氧化物酶和过氧化氢催化交联制备三维网状水凝胶,并用水凝胶负载碱性成纤维细胞生长因子(bFGF),制备SF-TG-bFGF水凝胶。对SF-TG-bFGF进行结构分析和形貌表征,检测SF-TG-bFGF对小鼠成纤维细胞L929增殖、迁移和胶原蛋白合成的影响。结果表明:SF-TG-bFGF水凝胶具有良好的力学性能、吸湿保湿性能和生长因子缓释性能;SF-TG-bFGF呈网状微观结构,弹性模量为21 kPa,溶胀率高达700%,能在144 h内缓释bFGF;SF-TG-bFGF能促进L929细胞的增殖、迁移和胶原蛋白合成,细胞存活率可达300%,细胞迁移率可达90%,胶原蛋白质量浓度可达4.5μg/mL。该研究制备的SF-TG-bFGF有望作为一种生物活性敷料应用于各类皮肤伤口治疗。To develop a new bioactive wound dressing,a horseradish peroxidase and hydrogen peroxide cross-linking system was used to catalyze the formation of a three-dimensional reticular hydrogel of silk fibroin(SF)and tyramine-modified gelatin(TG).This hydrogel was loaded with basic fibroblast growth factors(bFGF)and the SF-TG-bFGF hydrogel was formed.The SF-TG-bFGF hydrogel was structurally analyzed and morphologically characterized,and the effect of the SF-TG-bFGF hydrogel on the cell proliferation,migration capacity and collagen synthesis of mouse fibroblasts(L929)was examined.The results show that the SF-TG-bFGF hydrogel exhibits good mechanical properties,moisture absorption and moisture retention properties,and slow release performance of growth factors;SF-TG-bFGF has the network microstructure,the elastic modulus reaches 21 kPa,the swelling rate is as high as 700%,and bFGF is slowly released within 144 h;SF-TG-bFGF can promote the proliferation,migration and collagen synthesis of L929,with the cell survival rate,cell migration rate and mass concentration of collagen reaching 300%,90%and 4.5μg/mL,respectively.To sum up,the SF-TG-bFGF hydrogel prepared in this study is expected to be used as a bioactive dressing in the treatment of various skin wounds.
