摘 要:目的:麻醉剂的使用有利于减少实验动物在手术等操作中的痛苦,保障实验的顺利进行。本研究旨在阐明不同浓度和剂量的苯巴比妥钠对雌性BALB/c小鼠的麻醉效果,寻找适合实验手术的麻醉策略。方法:将不同浓度(2%、5%、10%)的苯巴比妥钠以75、100、125、150和200 mg/kg的剂量对雌性BALB/c小鼠进行腹腔注射。根据夹尾、前后肢退缩和眼睑反射等刺激指数进行麻醉评分。根据翻正反射的发生和持续时间记录各组的麻醉速度和麻醉持续时间。结果:苯巴比妥钠对雌性BALB/c小鼠的麻醉作用表现出明显的剂量依赖性,高剂量麻醉引起的呼吸抑制可能导致小鼠死亡。根据麻醉评分,当苯巴比妥钠用量大于或等于5%或200 mg/kg时,超过80%的小鼠能够达到手术所需的麻醉深度。2%浓度苯巴比妥钠处理小鼠达到手术麻醉深度的比率(达标率)分别为:75 mg/kg组0%,100 mg/kg组0%,125 mg/kg组50%,150 mg/kg组66.7%,200 mg/kg组100%;5%浓度苯巴比妥钠处理小鼠的达标率分别为:75 mg/kg组0%,100 mg/kg组0%,125 mg/kg组83.33%,150 mg/kg组100%,200 mg/kg组100%;10%浓度苯巴比妥钠处理小鼠的达标率分别为:75 mg/kg组50%,100 mg/kg组66.7%,125 mg/kg组100%,150 mg/kg组100%,200 mg/kg组100%。低浓度剂量的苯巴比妥钠能诱导镇静和催眠,高浓度剂量则诱导麻醉。其中,5%~125 mg/kg的苯巴比妥钠麻醉小鼠死亡率为0,麻醉诱导时间为(35.5±7.92)min,持续时间为(106±39.59)min;5%~150 mg/kg的苯巴比妥钠麻醉小鼠死亡率为0,麻醉诱导时间为(34.83±5.27)min,麻醉持续时间为(131.7±36.75)min。结论:苯巴比妥钠可以作为雌性BALB/c小鼠的全身麻醉剂,药物浓度和剂量都会影响麻醉效果。建议使用5%~125 mg/kg以及5%~150 mg/kg浓度-剂量组合的苯巴比妥钠来麻醉小鼠,并根据手术的时间要求调整麻醉剂的浓度、剂量。Objective:Anesthetics are of great importance in avoiding severe pain and suffering in animals and ensuring ex⁃perimental progress.This study was aimed at elucidating the anesthesia score of phenobarbital sodium as a general anesthetic at dif⁃ferent concentrations and doses in BALB/c mice,and finding the suitable anesthesia strategies for experimental surgeries.Methods:Phenobarbital sodium was administrated intraperitoneally at the doses of 75,100,125,150,and 200 mg/kg and randomly in different concentrations(2%,5%,and 10%)to female BALB/c mice.The anesthesia score was evaluated based on the stimu⁃lus index including tail⁃pinch,front and hind limb withdrawal,and eyelid reflexes.The speed and duration of anesthesia in different groups were recorded per the occurrence and duration of the righting reflex.Results:The anesthetic effect of phenobarbital sodium on female BALB/c mice showed an obvious dose⁃dependency.Respiratory suppression caused by high⁃dose anesthesia may lead to mouse death.Based on the anesthesia score,when the phenobarbital sodium treatment was greater than or equal to five percent or 200 mg/kg,more than 80%mice meet the anesthesia depth that surgical operation needed.The rates of achieving surgical anesthe⁃sia depth(standard⁃reaching rate)in mice treated with 2%sodium phenobarbital were 0%in the 75 mg/kg group,0%in the 100 mg/kg group,50%in the 125 mg/kg group,66.7%in the 150 mg/kg group,and 100%in the 200 mg/kg group.The standard⁃reaching rate of mice treated with 5%concentration of phenobarbital sodium were:0%in the 75 mg/kg group,0%in the 100 mg/kg group,83.33%in the 125 mg/kg group,100%in the 150 mg/kg group,and 100%in the 200 mg/kg group.The standard⁃reaching rate of mice treated with 10%concentration of phenobarbital sodium were:50%in the 75 mg/kg group,66.7%in the 100 mg/kg group,100%in the 125 mg/kg group,100%in the 150 mg/kg group,and 100%in the 200 mg/kg group.Se⁃dation and hypnosis were induced in the low⁃concentration dose group,and anesthesia was induced in the
