摘 要:目的通过检测静电纺丝技术制备的纳米银醋酸纤维素薄膜(cellulose acetate/silver nanoparticle,CA/AgNPs)、溴化十六烷基吡啶醋酸纤维素薄膜(cellulose acetate/cetylpyridine bromide,CA/CPB)、纳米银溴化十六烷基吡啶醋酸纤维素薄膜(cellulose acetate/silver nanoparticle/cetylpyridine bromide,CA/AgNPs/CPB)抗菌材料的生物安全性和抑菌作用,探讨新研发的抗菌口罩材料的应用价值。方法取CA/AgNPs、CA/CPB、CA/AgNPs/CPB,分别以MEM培养基及生理盐水为浸提介质,通过浸泡和滤菌器处理制备无菌浸提液。三种材料MEM培养基浸提液和MEM培养基分别培养小鼠成纤维细胞(NCTC clone 929,L929),应用MTT法检测材料的细胞毒性并于光学显微镜下观察细胞形态。脱脂棉浸泡生理盐水为阴性对照组,脱脂棉浸泡4%甲醛为阳性对照组,三种材料生理盐水浸提液为实验组,确定材料对新西兰兔皮肤的刺激性和原发性刺激指数。生理盐水为阴性对照,4%甲醛溶液为阳性对照,三种材料生理盐水浸提液为实验组,经皮内诱导、局部诱导、激发后观察豚鼠激发处皮肤情况,明确材料致敏性。采用琼脂扩散法和载体抑菌实验检测三种材料的抑菌效果。结果MTT检测结果,100%CA/AgNPs、CA/CPB、CA/AgNPs/CPB浸提液组在24 h内相对增殖率分别为97.26%、81.82%、89.67%,毒性均为1级。光学显微镜下,阴性对照组细胞为正常梭形或不规则三角形,各实验组细胞均有一定程度胞体细长的改变且随接触时间延长形态异常逐步加重。各材料组皮肤刺激和致敏试验结果均为阴性。CA/AgNPs、CA/CPB、CA/AgNPs/CPB各抑菌带≥0.5 mm,试样下无细菌繁殖且抑菌率均大于99.99%。结论CA/AgNPs、CA/CPB、CA/AgNPs/CPB均符合国标要求,具有良好的短期生物安全性和抑菌效果,有一定的应用前景。Objective To explore the application value of the newly developed antibacterial mask materials by testing the biosafety and antibacterial activity of nano-silver cellulose acetate film(celluloseacetate/silvernanoparticle,CA/AgNPs),cetylpyridinium acetate acetate film(celluloseacetate/cetylpyridinebromide,CA/CPB)and cetylpyridinium acetate acetate film(celluloseacetate/silvernanoparticle/cetylpyridinebromide,CA/AgNPs/CPB)prepared by electrospinning.Methods The aseptic extract of CA/AgNPs,CA/CPB,and CA/AgNPs/CPB were prepared by soaking and filter treatment with MEM medium and normal saline as extraction medium,respectively.Mouse fibroblasts(NCTCclone929,L929)were cultured in MEM medium extract and MEM medium,respectively.The cytotoxicity of the materials was detected by MTT method and the cell morphology was observed under light microscope.The group with defatted cotton soaked in normal saline was set as the negative control group,skimmed cotton soaked in 4%formaldehyde as the positive control group,and saline extract of three materials as the experimental group to determine the irritation and primary irritation index of the material to the skin of New Zealand rabbits.Normal saline was used as negative control,4%formaldehyde solution as positive control,and saline extract of three materials as experimental group.After intradermal induction,local induction and stimulation,the skin of guinea pigs was observed to clarify the allergenicity of the materials.Agar diffusion method and carrier bacteriostatic test were used to detect the bacteriostatic effect of the three materials.Results According to the results of MTT test,the relative proliferation rates of 100%CA/AgNPs,CA/CPB,and CA/AgNPs/CPB extract groups within 24 hours were 97.26%,81.82%,and 89.67%,respectively,and the toxicity was grade 1.Under light microscope,the cells in the negative control group were normal fusiform or irregular triangle,the cells in each experimental group had a certain degree of slender cell body change,and the abnormal morphology graduall
