摘 要:为了探究骆驼胎盘提取物(camel placenta extract,CPE)的免疫作用及其对巨噬细胞极化的影响,本研究将60只BALB/c小鼠随机分为空白对照组、模型对照组、阳性对照组和试验组。试验组小鼠又分为CPE高、中、低剂量(1.2、0.6、0.3 mg/mL)组,腹腔注射CPE连续14 d。除空白对照组外,其余组小鼠试验结束前7 d注射环磷酰胺致免疫抑制。通过ELISA、流式细胞术、组织病理学、荧光定量PCR和Western blot方法分别检测各组小鼠血浆IgG、细胞因子、淋巴细胞亚群、淋巴器官形态学变化以及腹腔巨噬细胞表面分子iNOS、CD206的mRNA和蛋白表达水平,以评估CPE对小鼠免疫功能和巨噬细胞极化的影响。结果显示,试验组小鼠的脾脏和胸腺指数,CD19^(+)、CD40^(+)细胞百分比和IgG水平均显著高于模型对照组(P<0.05);与模型对照组相比,试验组CD4^(+)、CD8^(+)细胞百分比和IL-4水平显著升高(P<0.05),而IL-2水平极显著降低(P<0.01);试验组巨噬细胞CD206 mRNA和蛋白表达水平均显著升高(P<0.05),而iNOS的表达无显著变化。以上结果表明,CPE对小鼠T、B细胞功能以及抑炎表型巨噬细胞极化均具有促进作用。In order to explore the effect of camel placenta extract(CPE)on immune function and macrophage polarization,60 BALB/c mice were obtained and randomly divided into the control group,the model group and the experimental group.The mice of the experimental group were further divided into the high,middle and low dose(1.2 mg/mL,0.6 mg/mL and 0.3 mg/mL)groups receiving injection by CPE intraperitoneally every day for 14 days.On the 7th day,all the groups,except the control group,were injected with cyclophosphamide for immunosuppression.The variations in plasma IgG,cytokines,lymphocyte subsets and lymphoid organs were detected by ELISA,flow cytometry and histopathology.Meanwhile,to evaluate the effect of CPE on the macrophage polarization in the mice,the mRNA and protein expression of iNOS and CD206 were detected by Q-PCR and Western blot,respectively,and the immunoenhancement effect of CPE and its possible mechanism were evaluated and analyzed.The results showed that the spleen and thymus indexes,the percentage of CD19^(+),and the CD40^(+)and IgG levels in the experimental group were significantly higher than those in the model group(P<0.05).Compared with the model group,the percentages of CD4^(+)and CD8^(+)and the IL-4 levels in the experimental group were significantly increased(P<0.05),but their IL-2 levels were significantly decreased(P<0.05).The expression of CD206 mRNA and the protein in the macrophages in the experimental group was significantly increased(P<0.05),but the expression of iNOS presented no significant changes.These results suggested that CPE might promote the immune function of T and B cells and the polarization of macrophages to M2 type(anti-inflammatory phenotype)in mice.
