摘 要:目的:研究双氢睾酮(DHT)对小鼠海马神经元细胞系(HT22)的影响,分析雄激素对小鼠海马神经元细胞的影响。方法:通过体外培养小鼠海马神经元细胞,用不同浓度的双氢睾酮诱导小鼠海马神经元细胞。24 h后倒置相差显微镜观察海马神经元细胞的生长密度与细胞形态特点。结果:加入含有不同浓度DHT的10%胎牛血清、100 U/mL青霉素和100μg/mL链霉素的DMEM培养基后,培养24 h后,对照组细胞编织状排布,生长密度约为100%;处理组1(0.25 ng/mL DHT)细胞呈编织状排列,重叠较少,连接密集,生长密度约为80%;处理组2(DHT浓度分为0.5 ng/mL)细胞较为紧密,生长密度约为70%;处理组3(1 ng/mL DHT)细胞大部分编织状连接,生长密度约为70%;处理组4(2 ng/mL DHT)细胞大部分互相连接,生长密度约为70%;处理组5(4 ng/mL DHT)可见少数连接的细胞,生长密度约为50%。结论:高剂量双氢睾酮在体外实验中影响小鼠海马神经元细胞的增长。Objective:To investigate the effect of dihydrotestosterone(DHT)on mice hippocampal neuron cells and to analyze the effect of androgen on mouse hippocampal neuron cells.Method:Mice hippocampal neurons were cultured in vitro induced by different concentrations of DHT.The growth density and morphological characteristics of hippocampal neurons were observed by inverted phase contrast microscope after 24 h.Results:In the control group the cers were cooven,and the growth density was about 100%.In treatment group 1(DHT concentration was 0.25 ng/mL),the cells were arranged in a braid pattern with less overlap and dense connections,and the growth density was about 80%.In treatment group 2(DHT concentration was 0.5 ng/mL),and the growth density was about 70%.In treatment group 3,most of the cells were braided and the growth density was about 70%.In treatment group 4,most of the cells were interconnected and the growth density was about 70%.In treatment group 5(DHT concentration was divided into 4 ng/mL),a few connected cells were seen,the growth density was about 50%.Conclusion:High dose of dihydrotestosterone can affect the growth of mice hippocampal neurons in vitro.
