基于代谢组学的狭叶锦鸡儿根醇提物抗类风湿性关节炎作用机制

摘　　要：目的:明确狭叶锦鸡儿根醇提物对类风湿性关节炎(rheumatoid arthritis,RA)的影响,探索狭叶锦鸡儿根醇提物抗RA的作用机制。方法:采用Ⅱ型胶原蛋白建立大鼠RA模型,通过病理组织切片评估狭叶锦鸡儿根醇提物对RA的影响;利用液相色谱-质谱联用技术(liquid chromatograph-mass spectromete,LC-MS)代谢组学技术分析狭叶锦鸡儿根醇提物对RA模型大鼠尿液代谢物的调控作用及代谢调控途径。结果:狭叶锦鸡儿根醇提物可改善RA模型大鼠足关节组织病理变化。通过LC-MS共鉴定出22种与RA相关的差异代谢物,狭叶锦鸡儿根醇提物能显著回调其中的19种。狭叶锦鸡儿根醇提物治疗RA大鼠涉及7条代谢通路,47种代谢物。结论:狭叶锦鸡儿根醇提物主要通过酪氨酸代谢、甘油磷脂代谢、半乳糖代谢等通路起到防治RA作用的。该研究阐明了狭叶锦鸡儿防治RA的作用机制,为狭叶锦鸡儿的临床合理应用提供参考。OBJECTIVE To clarify the effect of the ethanol extract of Caragana stenophylla roots on Rheumatoid arthritis(RA),and explore the mechanism of the ethanol extract of Caragana stenophylla roots against RA.METHODS TypeⅡcollagen was used to establish the rheumatoid arthritis model in rats,and the effect of ethanol extract of Caragana stenophylla roots on rheumatoid arthritis was studied by pathological tissue.LC-MS metabonomics technique was conducted on urine samples to analyze the regulation effect and metabolic regulation pathway of ethanol extract of Caragana stenophylla roots in rheumatoid arthritis model rats.RESULTS The ethanol extract of Caragana stenophylla roots improved the pathological changes of foot joints in rheumatoid arthritis model rats.LC-MS identified 22 metabolites related to rheumatoid arthritis,and the ethanol extract of Caragana stenophylla roots significantly decreased 19 of them.After treatment,47 metabolites in rheumatoid arthritis rats changed,involving 7 metabolic pathways.CONCLUSION The ethanol extract of Caragana stenophylla roots treat RA mainly by regulating tyrosine metabolism,glycerophospholipid metabolism,galactose metabolism and other pathways.This study not only provided the basis for elucidating the mechanism of Caragana stenophylla roots in preventing rheumatoid arthritis,but also provided reference for the rational clinical application of Caragana stenophylla roots.
