摘 要:【目的】研究叶面喷施水杨酸和冠菌素对当归早期抽薹过程中抗氧化酶、内源激素、多胺、抽薹相关基因表达量的影响,以期为更好解决当归早期抽薹问题提供理论依据。【方法】以喷施水杨酸和冠菌素的当归叶片为研究对象,通过分光光度法、超高效液相色谱串联质谱法、实时荧光定量PCR等实验手段分别测定抗氧化酶活性,内源激素、多胺含量及抽薹相关基因表达量,并通过正交偏最小二乘法分析以上指标与抽薹率的相关性。【结果】相较于对照,水杨酸和冠菌素处理均增强了当归过氧化氢酶和过氧化物酶活性,提高了茉莉酸、生长素、细胞分裂素、褪黑素含量,降低了丙二醛、脱落酸、腐胺、酪胺、苯乙胺含量,下调了抽薹正调控基因HD3A、FT、MADS8、AGL8、GA20X6、GA20OX1表达量,当归早期抽薹率下降。【结论】外源水杨酸和冠菌素均可抑制当归早薹;相较于水杨酸,冠菌素主要通过增强过氧化氢酶活性、降低内源脱落酸含量,抑制当归早薹;证明了水杨酸和冠菌素可以缓解早春干旱、低温、长日照等环境条件造成的当归抽薹。【Objective】To study the effects of foliar application of salicylic acid and coronatine on the ex⁃pression of antioxidant enzymes,endogenous hormones,polyamines and bolting-related genes in the early bolting process of Angelica sinensis,in order to provide a theoretical basis for better solving the early bolting problem of A.sinensis.【Method】Antioxidant enzyme activity,endogenous hormone,polyamine and rela⁃tive expression of bolting-related genes were determined by spectrophotometry,ultra-high performance liq⁃uid chromatography tandem mass spectrometry and real-time fluorescence quantitative PCR,respectively.The correlation between the above indexes and bolting was analyzed by orthogonal partial least squares method.【Result】Compared with the control,treatment with salicylic acid and coronatine increased the activities of catalase and peroxidase;increased the levels of jasmonic acid,auxin,cytokinin and melatonin;de⁃creased the levels of malondialdehyde,abscisic acid,putrescine,tyramine and phenylethylamine;downregu⁃lated the expression of HD3A,FT,MADS8,AGL8,GA20X6 and GA20OX1;and decreased the early bolt⁃ing rate of A.sinensis.【Conclusion】Treatment with exogenous salicylic acid and coronatine could inhibit early bolting of A.sinensis.Compared with salicylic acid,coronatine inhibited early bolting mainly by in⁃creasing catalase activity and reducing endogenous abscisic acid content.It was demonstrated that salicylic acid and coronatine could alleviate the bolting of A.sinensis caused by environmental conditions such as drought,low temperature and long sunshine in early spring.
