摘 要:目的观察当归芍药散对糖尿病肾损伤模型大鼠的干预作用。方法70只SD大鼠随机选取8只作为正常组,其余大鼠采用高糖高脂饲料喂养联合链脲佐菌素腹腔注射诱导糖尿病大鼠模型,造模成功后随机分为模型组、雷公藤多苷组(6 mg/kg)、当归芍药散低、中、高剂量组(7、14、21 g/kg)。给药8 w后,取其肾脏行免疫组化及Western印迹实验,测定肾组织E-钙黏蛋白(cadherin)、α-平滑肌肌动蛋白(SMA)及波形蛋白(Vimentin)表达。结果免疫组化及Western印迹实验显示,与模型组比较,正常组、当归芍药散中、高剂量组及雷公藤多苷组E-cadherin蛋白表达显著升高,Vimentin蛋白表达显著降低;正常组、当归芍药散高剂量组及雷公藤多苷组α-SMA水平显著降低(P<0.05,P<0.01)。Western印迹检测结果可见,与模型组比较,当归芍药散中剂量组α-SMA蛋白水平显著下调(P<0.01)。结论当归芍药散可能通过调节肾小管上皮细胞转分化减轻糖尿病肾病大鼠肾损伤。Objective To observe the intervention effects of Danggui Shaoyao Powder on diabetic nephropathy model rats.Methods Eight of 70 SD rats were randomly selected as the normal group,and the other rats were fed with high-sugar and high-fat diet combined with streptozotocin intraperitoneal injection to induce diabetic rat model.After successful modeling,the rats were randomly divided into model group,tripterygium glycoside group(6 mg/kg),Danggui Shaoyao Powder low,medium and high dose groups(7,14,21 g/kg).The expressions of E-cadherin,α-smooth muscle actin(SMA)and Vimentin in kidney tissue were determined by immunohistochemistry and Westem blot after 8 w administration.Results Immunohistochemistry and Western blot showed that compared with the model group,the expressions of E-cadherin protein were significantly increased and the expressions of Vimentin protein were significantly decreased in the normal group,the medium and high dose groups of Danggui Shaoyao Powder and the tripterygium glycoside group;the levels ofα-SMA in normal group,Danggui Shaoyao Powder high dose group and tripterygium glycoside group were significantly decreased(P<0.05,P<0.01).Westem blotting showed that compared with the model group,the level ofα-SMA protein in Danggui Shaoyao Powder medium dose group was significantly down-regulated(P<0.01).Conclusions Danggui Shaoyao Powder might reduce renal injury in rats with diabetic nephropathy by regulating the transdifferentiation of renal tubular epithelial cells.
