摘  要:为明确楚雄腮扁叶蜂雌虫头部交配响应相关基因表达情况,取交配后1、6和24 h(交配后24 h开始产卵)以及未交配的雌虫头部进行转录组分析,通过基因功能注释和富集分析明确头部交配响应基因表达情况,并预测可能涉及的生物学过程。结果表明,与未交配组相比,交配组在交配后1、6和24 h分别有621(448个上调、173个下调)、918(389个上调、529个下调)和172个(124个上调、48个下调)差异表达基因(DEGs)。交配后1 h上调DEGs共富集到115条GO(基因本体)术语/KEGG(京都基因与基因组百科全书)通路,多涉及物质和能量代谢,如氨基酸和脂类的合成与分解等;交配后6和24 h上调DEGs共富集到35条GO术语/KEGG通路,涉及范围较广,包括环境信号、细胞凋亡、生长发育及消化系统等。交配后1、6和24 h下调DEGs共富集到89条GO术语/KEGG通路,所涉及的范围更广,如疾病、循环系统、寿命、免疫和环境信号等。此外还找到31个神经肽/神经肽受体编码基因[包括1个DH⁃PBAN(与滞育和性信息素合成有关)同源序列]、78个OBPs(气味结合蛋白)以及1个SPR(性肽受体),但其中除了2个OBPs相关基因在交配后6 h显著下调表达外,其余基因的表达并未受到交配的影响。采用荧光定量PCR方法对15个DEGs的表达进行验证,结果与RNAseq分析的结果相一致。由此可见,交配引起了楚雄腮扁叶蜂雌虫头部基因表达的快速响应。To elucidate the expression of mating response⁃related genes in the head of Cephalcia chuxiongica females,the head of fe⁃males at 1,6 and 24 h after mating(females started to lay eggs at 24 h after mating)and unmated females were analyzed by RNAseq.Through functional annotation and enrichment analysis of differentially expressed genes(DEGs),the genes related to mat⁃ing response and their relevant biological functions were predicted.The results showed that compared with the unmated females,there were 621(448 up⁃regulated,173 down⁃regulated),918(389 up⁃regulated,529 down⁃regulated)and 172(124 up⁃regula⁃ted,48 down⁃regulated)DEGs in the mated females at 1,6 and 24 h after mating,respectively.At 1 h after mating,the up⁃regula⁃ted DEGs were enriched in 115 gene ontology(GO)terms/Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways,mostly involving in material and energy metabolism,such as the synthesis and catabolism of amino acids and lipids;the up⁃regulated DEGs at 6 and 24 h after mating were enriched in 35 GO terms/KEGG pathways,which involved in a wide range of biological processes,including environmental signal,apoptosis,development and digestive system,etc.On the other hand,the down⁃regulated DEGs at 1,6 and 24 h after mating were enriched in 89 GO terms/KEGG pathways,involving in an even wider range of biological processes,such as disease,circulatory system,longevity,immunity and environmental signals,etc.In addition,another 31 neuropeptide/neu⁃ropeptide receptor coding genes including a homologous sequence related to diapause hormone and pheromone biosynthesis activating neuropeptide(DH⁃PBAN),78 genes related to odorant binding proteins(OBPs)and 1 gene related to sex peptide receptor(SPR)were found.However,most of these genes were not affected by mating except that 2 OBPs⁃related genes were significantly down⁃reg⁃ulated at 6 h after mating.The expressions of 15 DEGs were further verified by fluorescence quantitative PCR,and the results were consistent with those of RNAse
