摘 要:目的比较红天麻、乌天麻及其差异成分的镇痛作用,为天麻道地药材的开发和临床应用提供参考。方法采用超高效液相色谱-串联质谱法(UPLC-MS/MS)对红天麻和乌天麻中19个酚酸类成分进行定量分析,比较其含量差异;使用热板致痛和醋酸致痛2种模型评估红天麻、乌天麻及其差异成分的镇痛作用。结果热板致痛和醋酸致痛实验结果表明,与空白对照组比较,红天麻可显著提高小鼠的痛阈值,红天麻和乌天麻均具有一定的镇痛作用;红天麻和乌天麻的差异成分琥珀酸、L-焦谷氨酸可显著提高小鼠的痛阈值,丁香酸、天麻素、琥珀酸、L-焦谷氨酸均具有一定的镇痛作用,其中琥珀酸、L-焦谷氨酸的镇痛作用更佳。结论与乌天麻相比,红天麻具有较强的镇痛作用,二者的差异成分琥珀酸和L-焦谷氨酸显示出较好的镇痛效果,可能是其镇痛作用差异的关键成分。Objective To compare the analgesic effects of G.elata BL.f.elata and G.elata BL.f.glauca and their different components;To provide reference for the development and clinical application of Gastrodia elata BL.Methods Totally 19 phenolic acids in Gastrodia elata BL.were quantified by high-performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS),and the difference of contents was compared.On this basis,the analgesic effects of G.elata BL.f.elata and G.elata BL.f.glauca were evaluated by hot-plate and acetic acid models.Results The experiment results of hot-plate and acetic acid models showed that compared with the blank control group,G.elata BL.f.elata could significantly increase the pain threshold in mice,and both G.elata BL.f.elata and G.elata BL.f.glauca had certain analgesic effects;succinic acid and L-pyroglutamate,the different components of G.elata BL.f.elata and G.elata BL.f.glauca,could significantly increase the pain threshold in mice,while syringic acid,gastrodin,succinic acid and L-pyroglutamate had certain analgesic effects,the analgesic effect of succinic acid and L-pyroglutamate was better.Conclusion Compared with G.elata BL.f.glauca,G.elata BL.f.elata has stronger analgesic effect,and succinic acid and L-pyroglutamic acid,the different components of G.elata BL.f.elata and G.elata BL.f.glauca,have better analgesic effect,which may be the key component of the difference in analgesic effect of G.elata BL.f.elata and G.elata BL.f.glauca.
