摘  要:目的研究唐古特大黄甲醇提取物及其不同极性部位的指纹图谱与体外抗炎作用的相关性,为揭示其抗炎作用物质基础提供依据。方法采用HPLC建立唐古特大黄甲醇提取物及其不同极性部位的指纹图谱,采用脂多糖刺激RAW264.7小鼠巨噬细胞制备炎性细胞模型,通过检测RAW264.7细胞一氧化氮(NO)分泌情况,考察唐古特大黄甲醇提取物及其不同极性部位的抗炎活性,采用偏最小二乘回归法(PLSR)建立谱效关系。结果PLSR结果表明,30个特征峰中有7个峰与抗炎率呈正相关(偏回归系数>0),23个色谱峰与抗炎率呈负相关(偏回归系数<0),其中1、11、27(大黄酸)、12、28(大黄素)、10、14、23(芦荟大黄素)、22、30号峰(大黄素甲醚)的变量重要性投影(VIP)>1。结论大黄酸、大黄素、芦荟大黄素、大黄素甲醚等对唐古特大黄抗炎药效的贡献最大。初步表明唐古特大黄的抗炎药效物质主要集中在二氯甲烷部位,抗炎作用是这些成分共同作用的结果。Objective To study the correlation between the fingerprints of methanol extract of Rheum tanguticum Maxim and its different polar parts and its anti-inflammatory effect in vitro;To provide the basis for revealing the substance basis of its anti-inflammatory effect.Methods The fingerprints of methanol extract and its polar parts were established by HPLC.The macrophages of RAW264.7 mice were stimulated by lipopolysaccharide to prepare inflammatory cell model.The secretion of nitric oxide(NO)in RAW264.7 cells was detected.The anti-inflammatory activity of methanol extract from Rheum tanguticum Maxim and its polar parts were investigated.Partial least squares regression(PLSR)was used to establish the spectral effect relationship.Results PLSR results showed that 7 of the 30 characteristic peaks were positively correlated with the anti-inflammatory rate(partial regression coefficient>0),and 23 chromatographic peaks were negatively correlated with the anti-inflammatory rate(partial regression coefficient<0).The VIP values of peaks 1,11,27(rhein),12,28(emodin),10,14,23(aloe emodin),22,30(physcion)were greater than 1.Conclusion The rhein,emodin,aloe emodin and physcion have the greatest contribution to the anti-inflammatory effect of Rheum tanguticum Maxim.It preliminarily proves that the antiinflammatory substances of Rheum tanguticum Maxim mainly concentrate in the dichloromethane part,and the antiinflammatory effect is the result of the joint action of these components.
