摘 要:采用原地共生萌发技术获得了鹤顶兰的原球茎,从中分离和筛选出了一株适合的共生菌——胶膜菌(HDL-2)。为验证分离菌株对鹤顶兰种子萌发的有效性,将HDL-2菌株与鹤顶兰种子在燕麦培养基上进行共生萌发试验。经过大约110 d的培养,HDL-2菌株可明显促进鹤顶兰种子萌发、形成原球茎并最终分化成幼苗,平均萌发率为(86.69±3.19)%,与无菌萌发相比有较高的萌发效率。表明分离得到的胶膜菌能促进鹤顶兰的种子萌发和原球茎发育。鹤顶兰种子实现共生萌发对于鹤顶兰资源的种质保育、人工栽培和可持续利用均具有重要意义。Using in situ symbiotic germination technology to obtain the protocorm of P.tancarvilleae,from which one highly compatible fungus(HDL-2)strain,Tulasnella sp.was isolated and screened out.In order to verify the effect of the isolated fungal strains on seed germination,seeds of P.tancarvilleae were co-cultured with the HDL-2 strain in an oatmeal agar medium.The HDL-2 enhanced seed germination and promoted protocorm growth and finally differentiated into seedlings after 110 days,and the average germination rate was(86.69±3.19)%.Compared with asymbiotic seed germination,seed germination promoted by HDL-2 had higher germination rate.Results showed that the isolated fungal strain HDL-2 could promote seed germination and protocorm growth of P.tancarvilleae.The success of symbiotic germination of P.tancarvilleae was of great importance for its germplasm conservation,artificial production and sustainable utilization.
