摘  要:目的通过检测基质金属蛋白酶-9(matrix metalloprotein-9,MMP-9)基因的rs3918242位点多态性,了解该位点相关基因型和等位基因在不同地区的分布特征,并探究其与梅州地区客家人群冠状动脉粥样硬化性心脏病(冠心病)发病风险之间的关系。方法选取梅州地区客家人群共79例确诊冠心病且无其他心脏疾病(瓣膜病、心肌炎、心肌病等)的患者进入冠心病组,对照组则选取同时期189例临床检查无冠心病客观证据且无其他共患病(瓣膜病、心肌炎、心肌病等)的体检人群,进行MMP-9基因的rs3918242位点多态性检测,分析该位点的基因型和等位基因及其他影响因素在两组间的相互关系、对比差异。结果本研究得到MMP-9基因的单核苷酸多样性(single nucleotide polymorphism,SNP)位点rs3918242的C等位基因频率高达89.74%,等位基因C和T频率在冠心病与体检健康人对比中没有显著性差异。在本研究人群中,存在CC、CT和TT三种基因型,CC最为常见。在冠心病组中,未检出TT。在基因型对比检验中,CT在P=0.1的水平上显现了更高的冠心病患病概率趋势。男性、高龄是冠心病发病的危险因素。MMP-9基因rs3918242相关的不同基因型组之间不存在年龄和性别间的差异。经对比发现广东梅州地区汉族客家人群与吉林长春地区汉族人群的rs3918242位点分布情况没有显著地域差异。结论本研究证明了在梅州地区客家人群中,MMP-9基因rs3918242位点存在两种等位基因C和T,冠心病与体检健康人群在等位基因类型方面没有显著性差异;但在基因型对比检验中,CT在P=0.1的水平上显现了更高的冠心病患病概率趋势。广东梅州地区与吉林长春地区的rs3918242位点分布情况没有显著地域差异。Objectives The rs3918242 polymorphism of the matrix metalloprotein-9(MMP-9)gene was detected to obtain the regional distribution characteristics of the related genotypes and alleles,and to understand the relationship between the polymorphism of the site and the incidence of coronary artery disease in the Hakka population of Meizhou area.Methods A total of 79 patients with confirmed coronary heart disease and no other heart diseases(valvular disease,myocarditis,cardiomyopathy,etc.)from the Hakka population in Meizhou were selected to form coronary heart disease group,while 189 physically examined individuals with no objective evidence of coronary heart disease and no other coexisting diseases(valvular disease,myocarditis,cardiomyopathy,etc.)during the same period were selected as control group.The rs3918242 polymorphism of the MMP-9 gene was detected and the relationships between the genotype and allele of the site and other influencing factors between the two groups were analyzed and compared for differences.Results In this study,the frequency of the C allele of single nucleotide polymorphism(SNP)site rs3918242 of the MMP-9 gene reached 89.74%,and there was no significant difference in the frequency of alleles C and T between coronary heart disease and non-coronary heart disease patients.In this study population,there were three genotypes:CC,CT,and TT,with CC being the most common in the coronary heart disease group and TT was not detected.In the genotype comparison test,CT showed a higher trend of coronary heart disease incidence probability at P=0.1 level.Male and old age were risk factors for coronary heart disease.There was no difference in age and gender between different genotypes related to MMP-9 rs3918242.After comparison,it was found that there was no significant regional difference in the distribution of rs3918242 locus between the Han Hakka population in Meizhou and the Han population in Changchun,Jilin.Conclusions This study proves that in the Hakka population in Meizhou region,there are two alleles,C
