摘 要:目的明确N7-甲基鸟苷(7-methylguanosine,m7G)相关基因对腹主动脉瘤(abdominal aortic aneurysm,AAA)的作用。方法从Gene Expression Omnibus(GEO)数据库中获取人类AAA和正常组织中m7G相关基因的表达数据,并进行差异分析。通过LASSO回归筛选与发病相关的基因并进行免疫浸润分析,根据其表达模式对AAA患者进行共识聚类分型,对两种亚型的差异基因进行基因本体论(gene ontology,GO)、京都基因和基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)分析。最后通过数据库预测筛选出的m7G相关基因的长链非编码RNA(long non-coding RNA,lncRNA)、微小RNA(microRNA,miRNA)。结果筛选出6个具有显著差异的m7G相关基因,其中IFIT5、NUDT10和NUDT4与AAA发病密切相关。这三者其可能影响CD4幼稚T细胞,自然杀伤(natural killer,NK)细胞,Tregs细胞的免疫浸润程度,根据其表达模式聚类的亚型与AAA的异质性密切相关,并构建了相关竞争性内源RNA(competing endogenouse RNA,ceRNA)网络。结论m7G相关基因IFIT5、NUDT10和NUDT4可能是治疗人类AAA的潜在靶点。Objectives To investigate the effect of 7-methylguanosine(m7G)methylation-related genes on abdominal aortic aneurysm(AAA).Methods Expression data of m7G related genes in human AAA and normal tissues were obtained from the Gene Expression Omnibus(GEO)database,and differential analysis was performed.LASSO regression was used to screen for genes related to the disease,and immune infiltration analysis was conducted.Based on their expression patterns,AAA patients were classified into consensus clusters,and differential genes between the two subtypes were subjected to gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)analysis.Finally,long non-coding RNA(lncRNA)and microRNA(miRNA)of m7G related genes were selected through database prediction.Results Six m7G related genes with significant differences were screened,among which IFIT5,NUDT10 and NUDT4 were closely related to AAA.These three genes may affect the degree of immune infiltration of CD4 naive T cells,natural killer(NK)cells and Tregs cells,and the clustered subtypes were closely related to the heterogeneity of AAA.A competing endogenouse RNA(ceRNA)network related to these genes was also constructed.Conclusions The m7G-related genes(IFIT5,NUDT10 and NUDT4)may be potential targets for the treatment of human AAA.
