摘 要:重组酶聚合酶扩增(RPA)技术是一种基于T4噬菌体复制机理为主要反应机理的核酸恒温扩增技术,广泛应用于动物疫病诊断、动物食品安全和遗传分析等领域。反应可在25~42℃温度条件下20 min完成快速扩增,具有条件温和、灵敏度高、特异性好、无需昂贵试验仪器和简便快捷等优点,适用于现场快速检测,已被应用于常见动物疫病的诊断。为让更多技术人员了解RPA技术诊断的优势,论文对RPA技术原理、引物探针设计、检测方法及在动物疫病诊断中的应用进行了分析。Recombinase polymerase amplification(RPA)technology is a nucleic acid constant temperature amplification technology based on the replication mechanism of T4 bacteriophage,which is widely used in animal infectious disease diagnosis,animal food safety,genetic analysis and other fields.The reaction can be rapidly amplified in 20 minutes under the lower temperature conditions of 25-42℃,with mild conditions,high sensitivity,good specificity,no need for expensive experimental instruments and simple and fast,suitable for point-of-care testing,has been used for the diagnosis of common animal infectious diseases,showing good sensitivity and specificity.In order to let more clinicians and scientific researchers understand the advantages of nucleic acid amplification diagnosis based on RPA technology,the principles of RPA technology,primer probe design,RPA detection methods,and applications,advantages,shortcomings and improvement directions in the diagnosis of animal infectious diseases were analyzed and summarized.
