桑寄生提取物对肾性高血压大鼠降压作用的初步研究

摘  要:目的 研究桑寄生提取物对肾性高血压大鼠(RHR)的降压作用及对肾功能的影响。方法 利用两肾两夹(2K2C)法制备大鼠肾性高血压(RH)模型。将SD雄性大鼠随机分为5组:假手术组,模型组,硝苯地平组,桑寄生组,桑寄生联合硝苯地平组(桑+硝组)。术后每周使用大鼠无创血压测量系统测量各组大鼠的血压,连续5 w,5 w后分别采用ELISA法、去蛋白终点微板法、二乙酰一肟比色法测定大鼠血浆肾素活性(PRA)、肌酐(Scr)、尿素氮(BUN)的含量。比较各组收缩压(SBP)及血浆中各指标的变化。结果 与假手术组相比,模型组大鼠SBP 1 w后开始明显升高(P<0.01)。与模型组相比,1 w后硝苯地平组和桑+硝组、2 w后桑寄生组SBP降低(P<0.05, P<0.01)。与硝苯地平组相比,桑+硝组在第5w降压作用有差异(P<0.05)。与假手术组相比,模型组Scr、BUN含量和PRA升高(P<0.05,P<0.01)。与模型组相比,硝苯地平组、桑寄生组、桑+硝组均能降低Scr、BUN的含量(P<0.05,P<0.01),桑寄生组、桑+硝组能降低PRA,硝苯地平组的PRA增高(P<0.05)。与硝苯地平组相比,桑寄生组的PRA明显下降(P<0.01),桑+硝组降低血Scr、BUN和PRA水平(P<0.05)。结论 桑寄生对RHR有降压作用,其机制可能与降低PRA有关,桑寄生能够降低Scr和BUN含量,对肾脏有保护作用,与硝苯地平联合后对肾脏的保护作用增强。Objective To investigate the antihypertensive effect of Taxilli Herba extract on renal hypertensive(RH)rats and its impact on renal function.Methods The two-kidney two-clip(2K2C)method was used to establish a rat model of RH.Male SD rats were randomly divided into five groups:a sham surgery group,a model group,a nifedipine group,a Taxilli Herba group,and a combination group(Taxilli Herba+nifedipine).The blood pressure of rats was measured weekly for five weeks using a non-invasive blood pressure measurement system after surgery.After five weeks,ELISA,deproteinization endpoint microplate method,and diacetyl monoxime colorimetric method were used to determine plasma renin activity(PRA),serum creatinine(Scr),and blood urea nitrogen(BUN)levels.Changes in systolic blood pressure(SBP)and plasma indicators were compared among the groups.Results Compared with the sham surgery group,the model group showed increased SBP one week later(P<0.01).Compared with the model group,the nifedipine group and the combination group showed SBP reduction one week later,and the Taxilli Herba group showed SBP reduction two weeks later(P<0.05,P<0.01).There was a difference in the antihypertensive effect between the nifedipine group and the combination group after five weeks(P<0.05).Levels of Scr,BUN,and PRA increased in the model group as compared with those in the sham surgery group(P<0.05,P<0.01).Compared with the model group,the nifedipine group,the Taxilli Herba group,and the combination group all reduced levels of Scr and BUN(P<0.05,P<0.01).The Taxilli Herba group and the combination group reduced PRA,whereas the nifedipine group increased PRA(P<0.05).Compared with the nifedipine group,the Taxilli Herba group showed decreased PRA(P<0.01),and the combination group decreased blood levels of Scr,BUN,and PRA(P<0.05).Conclusion Taxilli Herba has an antihypertensive effect on RH rats,which may be related to a decrease in PRA.Taxilli Herba can reduce Scr and BUN levels,thereby protecting the kidneys.Additionally,its protective effect on the ki
