摘 要:目的了解2022年湖州市甲型流感病毒的流行特点,明确流行株与疫苗株的匹配情况及流感病毒基因组进化特征。方法随机抽取2022年湖州市甲型流感病毒核酸阳性标本进行全基因组测序,采用Bioedit、MEGA 7.0软件对各基因片段进行系统发育分析及分子特征分析。结果2022年6月—12月甲型H3N2亚型流感病毒是湖州市流感的唯一流行型别。测得15株甲型H3N2亚型流感病毒全基因组,序列分析显示均属于3C.2a分支,为同一个进化来源,且与疫苗株A/Darwin/9/2021/H3N2关系较近。与疫苗株A/Darwin/9/2021/H3N2相比,15株H3N2亚型流感序列5个抗原决定簇位点的氨基酸序列与疫苗株成高度一致,HA蛋白上受体结合位点(RBS)的5个保守氨基酸也没有变化;HA蛋白增加了一个糖基化位点,NA蛋白糖基化位点没有变化。结论2022年湖州流行的甲型流感未发生具有流行病学意义的变异,接种当年度疫苗对甲型流感有一定的预防效果。需加强流感疫苗接种的宣传教育。Objective This paper aims to understand the epidemic characteristics of influenza A virus in Huzhou City in 2022,the matching of epidemic strains and vaccine strains,and the evolutionary characteristics of influenza virus genome.Methods The whole genome of influenza A virus positive samples in Huzhou City in 2022 was sequenced randomly,and the phylogenetic and molecular characteristics of each gene fragment were analyzed using Bioedit and MEGA 7.0 software.Results From June to December 2022,influenza A(H3N2)virus was the only epidemic type of influenza in Huzhou.The analysis for the whole genome of the 15 strains showed that they all belonged to the 3C.2a branch,closely related to the vaccine strain A/Darwin/9/2021/H3N2.Compared with the vaccine strain A/Darwin/9/2021/H3N2,the amino acid sequence of the five antigenic determinant sites of the 15 influenza strains remain unchanged,and the five conservative amino acids of the receptor binding site(RBS)on the HA protein also was the same;HA protein had an increased glycosylation site,while NA protein did not change its glycosylation site.Conclusion The influenza A does not have any variation with epidemiological significance,and vaccination in that year has a certain preventive effect on influenza A.It is necessary to strengthen the publicity and education of influenza vaccination.
