禽流感火麻油佐剂灭活疫苗实验室乳化条件研究

摘　　要：为了解决矿物油佐剂因不能在体内代谢而易引起免疫家禽发热、注射部位组织残留、形成肉芽肿等一系列副作用问题,寻找无副作用、无残留的新佐剂,采用药食同源植物油——火麻油作为佐剂,开展禽流感火麻油佐剂灭活疫苗实验室乳化条件研究,通过物理性状检验和无菌检验,确定疫苗的实验室制备工艺规程。结果表明,在油相中添加4.5%司班-80、在水相中添加3.0%吐温-80,油相与水相比例7∶3、乳化转速为10000 r/min的条件下疫苗乳化达到最佳效果。疫苗为微黄白色油包水型均匀乳剂,黏度合格,无细菌、霉菌污染,经3000 r/min离心15 min剂型稳定。初步说明火麻油可代替矿物油为佐剂制备出稳定的禽流感灭活疫苗。In order to solve a series of side effects caused by mineral oil adjuvants that cannot be metabolized in the body,such as fever of immune poultry,tissue residue at the injection site,and formation of granuloma due to the inability of mineral oil adjuvants to metabolize in the body.In the experiment,hemp seed oil,a homologous plant oil which has no side effect and no by-product,was applied as adjuvant.The laboratory emulsification conditions of inactivated vaccine of avian influenza oleoresin adjuvant were studied,and the laboratory preparation process of vaccine was determined through physical property test and sterility test.The results showed that the optimal emulsification conditions for the avian influenza inactivated vaccine with hemp seed oil adjuvant were 4.5%Span-80,3%Tween-80,an oil-to-water ratio of 7∶3,and emulsification at 10,000 r/min.It had a yellowish-white oily uniform emulsion appearance,qualified viscosity,and no bacterial or fungal contamination.The vaccine was stable with a dose of 3000 r/min centrifugation for 15 minutes.The preliminary results showed that hemp seed oil could replace mineral oil as adjuvant to prepare stable inactivated vaccine against avian influenza.
