摘  要:硫化氢(H_(2)S)和半胱氨酸(Cys)是许多生理过程的重要内源性调节剂,并且可以调节生物体内的氧化还原动态平衡的稳定。它们在体内浓度超过或低于人体所承受的范围时都有可能导致人体产生疾病。因此,对活细胞中的H_(2)S或Cys进行检测和成像是疾病诊断和治疗的迫切需求。与传统的检测H_(2)S或Cys方法相比,荧光探针的优势在于能够实时无误检测、对生物体损伤小、灵敏度高、选择性好和操作简便等优点。花菁结构包含多种,半花菁结构就是其中的一种。它拥有很多优点,比如摩尔吸光系数较大、吸收和发射波长相对较长、荧光量子产率较高、斯托克斯位移大等。尤其是近红外荧光探针(发射波长650~900 nm)因具有背景干扰低、对细胞损伤小、样品穿透性强、检测灵敏度高等优点,在疾病检测方面显示了良好的应用前景。我们通过半花菁结构与喹啉缩合,扩大其共轭结构得到近红外发射荧光团,进一步通过键合不同的识别基团设计合成了四种新型荧光探针,期望在生物体内实现实时检测H_(2)S或Cys的浓度异常的变化。Hydrogen sulfide(H_(2)S)and cysteine(Cys)are important endogenous regulators in many physiological processes,and can regulate the stability of the dynamic balance of redox in organisms.They may cause diseases when their concentration in the body exceeds or falls below the range that the human body can bear.Therefore,the detection and imaging of H_(2)S or Cys in living cells is an urgent need for disease diagnosis and treatment.There are many detection methods of H_(2)S or Cys,compared with traditional methods,fluorescent probes have the advantages of real-time error-free detection,little damage to organisms,high sensitivity,good selectivity and specificity,simple operation and so on,and are widely used in the detection of biomolecules including H_(2)S or Cys.There are many kinds of cyanine structures,including hemicyanine structure.It has many advantages,such as large molar absorption coefficient,relatively long absorption and emission wavelengths,high fluorescence quantum yield and large Stokes shift.Near-infrared(NIR)fluorescent probe(emission wavelength 650-900 nm)shows a good application prospect in disease detection because of its advantages of low background interference,little damage to cells,strong sample penetration and high detection sensitivity.We condensed the hemicyanine structure with quinoline to expand its conjugated structure.Then,we designed and synthesized four new types of NIR fluorescent probes by changing their recognition groups,hoping to detect the abnormal concentration changes of small biomolecules in human body in real time.
