摘 要:以中矮4号带芽茎段为试验材料,建立无菌组培苗,研究筛选组培苗继代增殖和生根培养的适宜培养基,建立中矮4号高效组培快繁技术体系。结果表明:中矮4号带芽茎段在适宜的初代培养基上获得了腋芽萌发的无菌组培苗。组培苗在增殖培养基MS+1.0 mg/L 6-BA+0.3 mg/L IAA+3.0 mg/L GA_(3)+30.0 g/L蔗糖上增殖生长较好,增殖系数为3.29,在该培养基上再添加200 mg/L纳米二氧化硅(NS),增殖效率可提高72.34%,增殖系数达5.67;组培苗在1/2MS+3.0 mg/L IBA+15.0 g/L蔗糖的培养基上暗培养5 d后,转移至1/2MS+1.0 g/L活性炭(AC)+15.0 g/L蔗糖的培养基中,生根效果最好,生根率达92%,平均单株根数为11.2条。In this study,the stem segments with buds of‘Zhong'ai 4'were used as experimental materials to establish sterile tissue culture seedlings,study and screen the suitable medium for subculture proliferation and rooting culture of tissue culture seedlings,and establish the efficient tissue culture and rapid propagation technology system of‘Zhong'ai 4'.The results showed that sterile tissue culture seedlings with axillary bud germination were obtained from the stem segments with buds of‘Zhong'ai 4'on suitable primary culture media.The tissue culture plantlets grew well on the proliferation medium MS+1.0 mg/L 6-BA+0.3 mg/L IAA+3.0 mg/L GA3+30.0 g/L sucrose,with a proliferation coefficient of 3.29.When 200 mg/L nano-silica(NS)was added to the medium,the proliferation efficiency could be increased by 72.34%,with a proliferation coefficient of 5.67;after dark culture for 5 days on the medium of 1/2MS+3.0 mg/L IBA+15.0 g/L sucrose,the plantlets were transferred to the medium of 1/2MS+1.0 g/L activated carbon(AC)+15.0 g/L sucrose,the rooting effect was the best,the rooting rate was 92%,and the average number of roots per plant was 11.2.
