摘　　要：目的分析三阴性乳腺癌(TNBC)患者中环状RNA circHIPK3(circHIPK3)的表达及对生物学行为的影响。方法选取2019年1月至2022年12月收治的三阴性乳腺癌(TNBC)患者124例和乳腺良性肿瘤(乳腺囊性增生和乳腺纤维腺瘤)患者50例。选取乳腺上皮细胞MCF-10A和TNBC细胞系BT-549、MDA-MB-231和MDA-MB-468。实时荧光定量PCR(qRT-PCR)检测TNBC组织、癌旁组织、MCF-10A和TNBC细胞中circHIPK3的表达。MDA-MB-468细胞分为空白对照组、circHIPK3过表达组、circHIPK3抑制组。检测细胞的增殖、侵袭、迁移能力及凋亡情况。结果TNBC组织中的circHIPK3相对表达量高于癌旁组织及乳腺良性肿瘤组织,癌旁组织高于乳腺良性肿瘤组织(P<0.05)。CircHIPK3在TNBC细胞中的相对表达量高于正常乳腺上皮细胞(P<0.05)。CircHIPK3过表达组转染48 h、72 h及96 h的增殖活性、细胞克隆数目和Edu/DAPI比值均高于空白对照组和circHIPK3抑制组,而circHIPK3抑制组低于空白对照组(P<0.05)。CircHIPK3过表达组的细胞侵袭数目及迁移率高于空白对照组和circHIPK3抑制组,而circHIPK3抑制组低于空白对照组(P<0.05)。CircHIPK3过表达组的凋亡率低于空白对照组和circHIPK3抑制组,而circHIPK3抑制组高于空白对照组(P<0.05);CircHIPK3抑制组的凋亡较空白对照组和circHIPK3过表达组更为明显,而circHIPK3过表达组凋亡更弱。结论CircHIPK3在TNBC患者中高表达,且能促进TNBC的增殖、迁移和侵袭,抑制凋亡,抑制circHIPK3表达TNBC的增殖、迁移和侵袭能力减弱,凋亡增加,可考虑将circHIPK3作为TNBC的治疗靶点。Objective To analyze the expression of circular RNA circHIPK3(circHIPK3)in patients with triple-negative breast cancer(TNBC)and its impact on biological behavior.Methods 124 patients with triple negative breast cancer(TNBC)admitted to the hospitalfrom January 2019 to December 2022 were selected.Breast epithelial cells MCF-10A and TNBC cell lines BT-549,MDA-MB-231 and MDA-MB-468 were selected.CircHIPK3 expression in TNBC tissues,paracancer tissues,MCF-10A and TNBC cell lines were detected by quantitative real-time PCR(qRT-PCR).MDA-MB-468 cells were divided into blank control group,circHIPK3 overexpression group and circHIPK3 inhibition group.The proliferation,invasion,migration and apoptosis of cells were detected.Results The relative expression level of circHIPK3 in TNBC tissue was higher than that in paracancer tissue and breast benign tumor tissue,and the paracancer tissue was higher than that in breast benign tumor tissue(P<0.05).The relative expression of circHIPK3 was higher in TNBC cell lines(BT-549,MDA-MB-231 and MDA-MB-468)than in normal mammary epithelial cells(MCF-10A)(P<0.05).The relative expression of circHIPK3 in TNBC cells was higher than that in normal mammary epithelial cells(P<0.05).The proliferative activity at 48 h,72 h and 96 h after transfection,the number of cell clones and Edu/DAPI ratio of circHIPK3 overexpression group were higher than those of blank control group and circHIPK3 inhibitory group,while those of circHIPK3 inhibitory group were lower than those of blank control group(P<0.05).Transwell and scratch assays showed that the cell invasion data and migration rate of the circHIPK3 overexpression group were higher than those of the blank control group and the circHIPK3 inhibition group,while the circHIPK3 inhibition group was lower than the blank control group(P<0.05).The apoptosis rate in the circHIPK3 over-expression group was lower than that in the blank control and circHIPK3 inhibition groups,while the circHIPK3 inhibition group was higher than that in the blank control group(P<0.
