摘  要:目的:探讨黄芪多糖通过神经调节因子-1(NRG-1)/表皮生长因子受体(ErbB)信号通路对心肌缺血再灌注损伤大鼠的影响。方法:无特定病原体(SPF)级健康雄性SD大鼠70只,随机分为对照组、模型组、黄芪多糖低浓度组、黄芪多糖高浓度组及阳性对照组,每组14只。对照组进行假手术,其余各组采用左前降支结扎建立心肌缺血再灌注损伤大鼠模型。造模前30 d,模型组、对照组和阳性对照组大鼠分别按1 mL/kg尾静脉注射生理盐水,术后每日1次注射;黄芪多糖低浓度组、黄芪多糖高浓度组分别按1 mL/kg尾静脉注射20、40 mg/mL黄芪多糖溶液,术后每日1次注射。造模前30 min,阳性对照组给予4μg/kg NRG-1溶液静脉注射,术后每日1次注射。各组均连续干预30 d。检测各组大鼠血清心肌酶水平、心肌组织细胞凋亡指数、心肌组织半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)和NRG-1、磷酸化ErbB2(p-ErbB2)、B淋巴细胞瘤-2(Bcl-2)、Bcl-2关联X蛋白(Bax)表达水平。结果:治疗后,与模型组比较,黄芪多糖低浓度组、黄芪多糖高浓度组及阳性对照组大鼠血清天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)、心肌钙蛋白T(cTnT)水平降低,心肌组织细胞凋亡指数降低,心肌组织Caspase-3阳性表达率降低,心肌组织NRG-1、p-ErbB2、Bcl-2表达水平升高,Bax表达水平降低,差异均有统计学意义(P<0.05),且黄芪多糖高浓度组以上指标的变化均优于黄芪多糖低浓度组,差异均有统计学意义(P<0.05)。结论:黄芪多糖可减轻心肌缺血再灌注损伤大鼠的心肌损伤,抑制心肌细胞凋亡,其作用可能与激活NRG-1/ErbB信号通路,调节凋亡相关蛋白Bcl-2、Bax、Caspase-3表达有关。Objective:To analysis the effect of astragalus polysaccharides on rats with myocardial ischemia-reperfusion injury based on neuregulin-1(NRG1)/epidermal growth factor receptor(ErbB)signaling pathway.Methods:A total of 70 healthy male SD rats with specific pathogen(SPF)grade were randomly divided into control group,model group,astragalus polysaccharide low concentration group,astragalus polysaccharide high concentration group,and positive control group,with 14 rats in each group.Sham operation was performed in the control group,and ligation of the left anterior descending branch was performed in the other groups to establish the rat model of myocardial ischemia-reperfusion injury.at thirty days before modeling,rats in model group,control group and positive control group were injected 0.9%NaCl by 1 mL/kg,once a day after surgery.The low concentration group,and high concentration group were injected 20 and 40 mg/mL of astragalus polysaccharide solution by 1 mL/kg,respectively,and were injected once a day after surgery.The positive control group was given intravenous injection of 4μg/kg NRG-1 solution at 30 min before modeling,and was injected once a day after surgery.All groups were continuously treated for 30 days.Serum myocardial enzyme levels,myocardial tissue apoptosis index,expression levels of cysteine aspartate protease-3(Caspase-3)and NRG-1,phosphorylated ErbB2(p-ErbB2),B-lymphoblastoma-2(Bcl-2),and Bcl-2 associated X protein(Bax)in myocardial tissue were detected in each group.Results:After treatment,compared with model group,serum levels of aspartate aminotransferase(AST),lactate dehydrogenase(LDH),creatine kinase isoenzyme(CK-MB),and cardiac calcin T(cTnT)of rats in low concentration of astragalus polysaccharide group,high concentration of astragalus polysaccharide group and positive control group decreased,and apoptosis index of myocardial tissue decreased.The positive expression rate of Caspase-3 in myocardial tissue decreased,the expression levels of NRG-1,p-ERBB2 and Bcl-2 in myocardial tissue increa
