摘 要:目的:观察半枝莲多糖(SBPS)不同给药方式对肝癌组织中血管生成的影响。方法:建立裸鼠肝癌模型,随机分为对照组及给药组(口服SBPS组、肝动脉灌注SBPS组),给药组分别灌胃或经肝动脉灌注SBPS提取物(100mg/kg),间隔1周给药1次,连续给药3次,末次给药24h后检测荷瘤大鼠抑瘤率,通过免疫组化分析肿瘤组织微血管密度(MVD)并经qPCR和Western blot方法检测肿瘤组织中血管内皮生长因子(VEGF)的mRNA和蛋白表达水平。结果:较对照组,给以SBPS后大鼠抑瘤率显著提高(P<0.05),且经肝动脉灌注方式高于灌胃组(P<0.01);MVD实验结果显示,较灌胃组,经肝动脉灌注SBPS提取物对肝癌组织中微血管的生长抑制作用更显著(P<0.01);此外,较灌胃组,经肝动脉灌注SBPS提取物更显著地降低VEGF的mRNA和蛋白水平(P<0.01)。结论:SBPS能显著抑制大鼠肝癌肿瘤的生长及肝癌组织血管生成,其机制可能与下调VEGF的mRNA和蛋白表达水平相关,且肝动脉灌注给药效果优于口服。Objective:To observe the effects of different administration modes of Scutellaria barbata polysaccharides(SBPS)on angiogenesis of rat hepatocellular carcinoma.Methods:A nude mouse liver cancer model was established and randomly divided into control group and treatment group(oral SBPS group and hepatic artery perfusion SBPS group).The SBPS extract(100mg/kg)was injected intragastric or hepatic artery in the administration groups,once every 1 week,for 3 consecutive times.The tumor inhibition rate of tumor-bearing rats was detected at 24h after the last administration.The tumor microvascular density(MVD)was analyzed by immunohistochemistry,and the mRNA and protein expression levels of vascular endothelial growth factor(VEGF)were detected by qPCR and Western blot.Results:Compared with the control group,the tumor inhibition rate of the rats was significantly increased after the administration of SBPS(P<0.05),and the mode of hepatic arterial perfusion was significantly higher than that of gavage group(P<0.01).The results of MVD experiment showed that compared with the intragastric group,transhepatic arterial infusion of SBPS extract had a more significant inhibitory effect on growth of microvessels in liver cancer tissues(P<0.01).In addition,compared with the intragastric group,transhepatic arterial infusion of SBPS extract significantly decreased mRNA and protein levels of VEGF(P<0.01).Conclusion:The SBPS can significantly inhibit the growth and angiogenesis of liver cancer in rats,and the mechanism may be related to the down-regulation of VEGF mRNA and protein expression levels,and the anti-tumor effect of hepatic artery perfusion is better than that of oral administration.
