摘 要:目的研究50 Hz磁场暴露对Neuro2a/APP(695)细胞基因转录的影响。方法Neuro2a/APP(695)细胞简单随机分为假性辐照组和磁场暴露组(n=3)。暴露条件为1.0 mT 50 Hz正旋波磁场,4 h/d,连续3 d;假性辐照组细胞除无磁场暴露外,余同磁场暴露组。暴露结束后按标准方法送样,进行转录组测序检测,包括样品RNA提取、数据质量评价和数据分析等。结果50 Hz磁场暴露后Neuro2a/APP(695)细胞有141种定性的基因表达发生了明显的变化,其中有129种表达上调、12种表达下调。GO富集分析结果显示,Neuro2a/APP(695)细胞中62种上调差异表达基因涉及432类显著GO功能富集,其中涉及生物学过程337类、细胞组分16类、分子功能79类;3种下调差异表达基因共涉及275类显著GO功能富集,其中涉及生物学过程260类、细胞组分5类、分子功能10类。KEGG通路分析显示,这些差异基因可能参与了20条KEGG通路,其中2个下调差异表达基因通路,18个上调差异表达基因通路。结论50 Hz磁场暴露可能对Neuro2a/APP(695)细胞的黏附连接、Hippo信号通路、糖胺聚糖生物合成-硫酸软骨素/硫酸皮肤素、蛋白消化和吸收和甘露糖型O-聚糖生物合成等方面产生影响,以上通路可能是50 Hz磁场对Neuro2a/APP(695)细胞产生影响的主要途径。Objective To investigate the impact of exposure to 50 Hz magnetic field on transcription in Neuro2a/APP(695)cells.Methods Neuro2a/APP(695)cells were divided into radiation group and sham radiation group(n=3).The cells in the former group were exposed to 50 Hz magnetic field(1 mT,4 h/d)for 3 consecutive days,and those from the latter group were only placed in the same coil system under the same conditions but without 50 Hz magnetic field.After exposure,all samples were collected to undergo standard procedures for transcriptome sequencing,including RNA extraction,data quality evaluation,and data analysis.Results There were 141 identified significant differentially expressed genes(DEGs)in Neuro2a/APP(695)cells after exposure,of which 129 were up-regulated and 12 were down-regulated.Gene Ontology(GO)enrichment analysis showed that the 62 up-regulated DEGs were involved in 432 significantly functional properties,of which 337 belong to biological processes,16 to cellular components and 79 to molecular functions.While,the 3 down-regulated DEGs were involved in 275 significantly functional properties,of which 260 belonged to biological processes,5 to cellular components and 10 to molecular functions.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis revealed that these DEGs may be involved in 20 differential KEGG pathways,including 2 down-regulated pathways and 18 up-regulated pathways.Conclusion Exposure to 50 Hz magnetic field may affect adhesion junction,Hippo signaling pathway,glycosaminoglycan biosynthesis-chondroitin sulfate/dermatan sulfate,protein digestion and absorption,mannose type O-glycan biosynthesis in Neuro2a/APP(695)cells,and these may be the main pathways that the exposure affects the cells.
