摘  要:目的测试连续电纺纳米纤维材料所生产的医用纱布的组织相容性和可降解性。方法分别将纳米纤维纱布与普通医疗纱布种植于Sprague-Dawley(SD)大鼠的背部皮下组织及腹膜下,以观察SD大鼠创面的痊愈时间、创面痊愈状况,以及组织中炎症反应状况和有机纤维的降解状况。结果植入了纳米纤维纱布和普通纱布填塞物大鼠的创面痊愈程度分别为甲级和乙级。在术后的第3天,对皮下取材和组织进行苏木精-伊红染色,结果表明:普通纱布的皮下组织结构已开始与黏膜、脂肪进行结合,其与组织并没有很好地结合;而纳米纤维纱布则是轮廓上清晰可见,一整片纱布单独存在,并不能与其他黏膜结合,且以纱布为核心的结构中出现了很密的细胞。同样,在HE染色结果中,普通纱布组表明玻璃纤维比较粗大,结构较稀疏,生物相容性不良,而纳米玻璃纤维的纱布组表明玻璃纤维和组织之间已成为比较紧密的结构。免疫组化结果表明在普通纱布的纤维组织中巨噬细胞染料为明显阳性,仍然有慢性炎症反应,而在纳米纤维纱布中巨噬细胞染料阳性但不明显,组织依然生长致密。结论与传统的医用纱布比较,纳米纤维纱布具有较好的相容性和可降解性。Objective To evaluate the biomedical histocompatibility and biodegradability of medical gauze made of continuous electrospun nanofiber materials.Methods Nanofiber gauze and common medical gauze were implanted into the back of SD rats under the skin and the peritoneum.The wound healing time,wound healing condition,inflammation in the tissue mass,and fiber degradation were observed.Results The wound of the rats implanted with nanofiber gauze and common gauze showed a healing grade of Grade A and Grade B,respectively.After three days,the samples of subcutaneous tissues were treated with hematoxylin-eosin staining(HE staining).The results indicated that:in the common gauze group,the subcutaneous tissue structure had begun to combine with mucosa and fat,but the gauze was not bonded well with the tissue;while in the nanofiber gauze group,it showed clearly visible contours,but a whole piece of gauze existed alone and was not bonded to mucosa.In addition,it showed dense cells with the gauze at the center.Regarding to the result of the HE staining,the common gauze group showed that the glass fibers were relatively coarse,with a sparse structure and poor biocompatibility,while the nanofiber group showed that the glass fiber and tissue had forged a relatively tight structure.The immunohistochemical results showed that macrophage staining was significantly positive in the fibrous tissue of common gauze,indicating that there was still a chronic inflammatory reaction,while the staining of macrophages in the nanofiber gauze was positive but not obvious,and the tissue grew densely.Conclusion Compared with traditional medical gauze,nanofiber gauze has better compatibility and degradability.
