摘  要:试验旨在探究紫花地丁香豆素类提取物体外对新城疫病毒的抑制作用。试验以紫花地丁为材料,制备紫花地丁香豆素提取物体,利用高效液相色谱法检测提取物中秦皮甲素和秦皮乙素的含量,利用CCK-8方法检测紫花地丁香豆素类提取物对新城疫病毒(NDV)感染鸡胚成纤维细胞的影响。结果表明,紫花地丁中秦皮甲素和秦皮乙素的含量分别为2.58、7.66 mg/g;紫花地丁香豆素类提取物在DF-1细胞中的半数致死浓度为185.2 mg/L。紫花地丁香豆素提取物浓度以剂量依赖的方式抑制NDV的复制,当提取物浓度为100 mg/L时,对NDV的复制抑制率最高,为74.31%。在NDV吸附抑制中,当紫花地丁香豆素类提取物作用浓度为100 mg/L时,对NDV吸附抑制率最高,为67.4%。研究表明,紫花地丁香豆素类提取物在体外具有明显的抗NDV感染作用。The experiment was to study the inhibitory effect of coumarin extract from Viola yedoensis Makino on Newcastle disease virus in vitro.The experiment used Viola yedoensis Makino as the material to prepare the coumarin extract,used high performance liquid chromatography to detect the content of aesculin and aesculetin in the extract,and used CCK-8 method to detect the effect of extract on Newcastle disease virus(NDV)infection of chicken embryo fibroblasts.The results showed that the contents of aesculin and aesculetin in Viola yedoensis Makino were 2.58,7.66 mg/g,respectively.The median lethal concentration of coumarin extract from Viola yedoensis Makino on DF-1 cells was 185.2 mg/L.Coumarin extract from Viola yedoensis Makino inhibits NDV replication in a dose-dependent manner.When the extract concentration was 100 mg/L,the replication inhibition rate against NDV virus was the highest with the value of 74.31%.In the inhibition of NDV adsorption,when the concentration of lycopene extract was 100 mg/L,the inhibition rate of NDV virus was the highest with the value of 67.4%.The study indicates that the coumarin extract from Viola yedoensis Makino has obvious anti-NDV effect in vitro.
