摘 要:为了深入探索采前外源多胺预处理对采后杏鲍菇子实体内源多胺、乙烯释放量的诱导及其常温贮藏品质的影响,以清水喷施为对照(CK),用0.5、1.0、2.0、4.0 mmol/L外源亚精胺(Spd)、精胺(Spm)对杏鲍菇进行采前喷施处理,测量菇柄及菇伞组织的可溶性糖及可溶性蛋白含量,筛选最适的Spd及Spm处理浓度;并研究了采前喷施最适浓度的Spd及Spm对采后杏鲍菇子实体内源多胺含量、乙烯释放量、S-腺苷蛋氨酸(SAM)活性、S-腺苷蛋氨酸脱羧酶(SAMDC)活性及细胞膜系统的影响。结果表明:外源Spd及Spm的最佳处理浓度均为1.0 mmol/L,该处理能保持子实体较高的可溶性糖及可溶性蛋白含量,维持菇体较好的贮藏表型;采前喷施外源Spd、Spm可促进子实体内源Spd、Spm的积累,并延缓丙二醛(MDA)及细胞膜相对透性的上升,维持较低的活性氧水平,一定程度抑制乙烯释放,同时降低乙烯释放量和内源多胺的比值(E/Spd和E/Spm),增强SAMDC活性水平,促使SAM更多地转向多胺合成方向,维持子实体较高的贮藏品质;在采后常温贮藏期间轻度的环境胁迫下,杏鲍菇子实体的SAM库容量相对充足,多胺与乙烯之间并不存在明显的拮抗效应。In order to explore the induction effects of preharvest treatment with exogenous polyamines on the endogenous polyamine contents,ethylene production and storage quality of postharvest P.eryngii,we determined the optimal concentration of spermidine(Spd)and spermine(Spm)though analyzing the soluble sugar and protein contents of P.eryngii treated with exogenous Spd and Spm at concentrations of 0,0.5,1.0,2.0 and 4.0 mmol/L before harvest,in which purified water spray served as a control(CK).Then,the content of endogenous Spd,Spm and S-adenosylmethionine(SAM),ethylene production,the activity of S-adenosylmethionine decarboxylase(SAMDC)and damage degree of cellular membrane system of postharvest P.eryngii sprayed with the optimal concentrations of Spd and Spm before harvest were investigated.The results indicated that the optimal concentrations for both Spd and Spm were 1.0 mmol/L.Compared with the control,the fruiting bodies treated with exogenous polyamines showed better storage quality with higher content of soluble sugar and protein.Preharvest polyamine applications could promote the accumulation of endogenous Spd and Spm,suppress the potentiation of malondialdehyde(MDA)and relative membrane permeability,maintain lower level of reactive oxygen species(ROS),and inhibit the release of ethylene,decrease the ratio of ethylene and polyamines(E/Spd and E/Spm),trigger the activity of SAMDC,and integrate more SAM turn to polyamines synthesis.Under the mild environmental stress of postharvest storage,SAM pool was relatively sufficient,and there was no significant antagonistic relationship between polyamines and ethylene.
