摘 要:为探究适合云实粗多糖色素、蛋白质等杂质的清除方法,该研究以多糖保留率、色素和蛋白质清除率的综合吸附效率为评价指标,对不同类型的大孔吸附树脂进行筛选,后采用单因素实验法,对样品浓度、初始pH值、吸附温度和过柱流速进行优化,利用优化条件对云实粗多糖进行纯化,并评价了树脂吸附后综合除杂效果;此外,还利用高效液相凝胶色谱和傅里叶变换红外光谱技术对纯化前后多糖部分特征进行了分析。结果表明:D101大孔吸附树脂对云实根粗多糖具有较好的综合纯化效率,在多糖浓度3.0 mg/mL,初始pH值7.0,吸附时环境温度30℃,流速2.0 BV/h时有最佳纯化效率,多糖保留率达(70.48±1.19)%,色素清除率为(26.26±3.53)%,蛋白质清除率为(56.65±1.90)%;多糖特征分析结果表明在吸附前后云实多糖糖链结构稳定;综上,利用D101大孔吸附树脂可高效纯化云实粗多糖,且条件温和,不涉及有机试剂,可用于云实根粗多糖的大量除杂。该研究也可为云实根纯多糖的制备提供参考。To explore more effective method to remove impurities such as pigments and proteins in crude polysaccharide from the root of Caesalpinia decapetala(Roth)Alston(CdPs),comprehensive adsorption effect index calculated from the values of the recovery ratio of carbohydrate(Drr%),decoloration(Dcr%)and deproteinization(Dpr%)ratio is used as the indicator to obtain optimal resin and adsorption conditions.The initial pH value,adsorption temperature,concentration of sample and sample flow rate were optimized by using single-factor experiments.And the comprehensive impurity removal effect of CdPs was evaluated after resin adsorption under the optimized conditions.In addition,HPGPC and FT-IR were used to analyze the characteristics of CdPs before and after purification.The results showed that D101 macroporous adsorption resin exhibited the best comprehensive adsorption effect.The Drr%,Dcr%and Dpr%were(70.48±1.19)%,(26.26±3.53)%and(56.65±1.90)%respectively,under optimized conditions as follows:the initial polysaccharide concentration of 3 mg/mL,the initial pH of 7,the temperature of 30℃,and the flow rate of 2.0 BV/h.The results of partial structural characteristic of CdPs showed that the polysaccharide chain structure was stable before and after adsorption.It can be seen that D101 macroporous adsorption resin can be used to purify efficiently the crude CdPs.The impurity removal conditions are mild,and no organic reagents are used,which can be used for large amounts of crude CdPs.This study can provide reference for purifying polysaccharides from the root of C.decapetala(Roth)Alston as well.
