摘 要:目的:对马鹿茸多肽A的表面结构进行分析,并探讨其结构对药效的作用。方法:采用原子力显微镜观察马鹿茸多肽A的表面结构。结果:马鹿茸多肽A表面呈三维的曲面、板状结构;其基部为不规则网格状结构,3D结构显示其网格边缘具有粗细不均、长短不等的支链形成椎体状突起,呈栅状或片状。质量浓度为0.50、0.25、0.10µg·mL^(–1)的马鹿茸多肽A的粗糙度分别为0.6672、0.7000和0.7900,表面高度分别为14.5105、39.4588、6.1397 nm。在相同波长下,一维各向同性功率谱密度越大,则二维各向同性功率谱密度越大,样品对于测定的响应越灵敏。结论:马鹿茸多肽A的曲面板状结构可增加与作用靶点的接触面积,其栅状或片状椎体支链结构则更有利于和作用靶点的相互接触或渗入而易发挥药效作用。随着质量浓度的增大,其粗糙度逐渐变小;而表面高度没有随质量浓度的变化呈现出规律性的变化。Objective:To analyze the surface structure of pilose antler polypeptide A and explore its effect on drug efficacy.Methods:The surface structure of pilose antler polypeptide A was analyzed by atomic force microscopy(AFM).Results:The surface of pilose antler polypeptide A shows a three-dimensional(3D)curved surface and plate structure.Its base is an irregular grid structure,and the 3D structure shows that its grid edge has branch chains of uneven thickness and length,so as to form vertebral body-like protrusions,which are in the form of grid or sheet.The roughness of pilose antler polypeptide A at 0.50,0.25,0.10µg·mL^(-1) are 0.6672,0.7000,and 0.7900,respectively.The surface heights are 14.5105,39.4588,6.1397 nm,respectively.At the same wavelength,a larger isotropic power spectral density indicates a larger two-dimensional isotropic power spectral density,and the sample is more sensitive to the determination.Conclusion:The curved surface and plate structure of pilose antler polypeptide A can effectively increase the contact area with the target,and its branch chains of vertebral body-like protrusions,which are in the form of grid or sheet,are more conducive to the mutual contact or infiltration of the target and easy to play the drug effect.With the increase in concentration,the roughness decreases gradually.The surface height does not change regularly with the concentration.
