摘 要:目的:用超高效液相色谱-四级杆串联飞行时间质谱法(UPLC-Q-TOF-MS)研究摔跤运动员比赛前后血清代谢变化特征,为科学化营养补充实现快速恢复以及发现训练监控新指标提供新思路。方法:筛选女子摔跤运动员11人(年龄22.09±2.77岁,身高1.63±0.04 m,体重60.45±8.43 kg,运动等级为一级运动员或运动健将),分别于比赛前1天、比赛结束归队后次日清晨7时空腹进行肘静脉采血,共计采血2次。每次各采集肘静脉血两管,一管(3 mL)用于全血生化指标分析,另一管(5 mL)离心后取血清用于生化指标检测和代谢组学检测。采用UPLC-Q-TOF-MS进行代谢组学检测及数据分析,确定运动前后的代谢差异物,获取的差异代谢物进行通路分析,确定比赛前后涉及的潜在代谢通路变化。同时对检测的生化指标与代谢物进行皮尔森相关性分析。结果:(1)运动员赛后血清尿素(BU)、血清总睾酮(TT)水平显著高于赛前(P<0.01,P<0.05),红细胞压积(HCT)显著低于赛前(P<0.05)。(2)根据组间比较的P值小于0.05和正交偏最小二乘分析的变量重要性(variable importance in projection,VIP)值大于1的标准,确定了42个代谢特征峰,筛选出19个与比赛相关的差异代谢物,其中13个代谢物赛后水平上调,6个代谢物赛后水平下降。(3)确定了2条比赛后出现异常的通路,分别为苯丙氨酸代谢通路,色氨酸、苯丙氨酸和酪氨酸合成通路。(4)发现血红蛋白(HGB)、HCT、BU、血清肌酸激酶(CK)、血清皮质醇(C)与多个代谢差异物存在显著相关,特别是溶血磷脂酰乙醇胺LPE(20:4)。结论:代谢组学分析法可以很好地揭示运动比赛代谢特征,为实现科学精准营养补充和筛选训练监控新指标提供新思路,摔跤运动员在比赛前期和赛后补充足够的蛋白质,特别是进行色氨酸、酪氨酸和支链氨基酸的针对性营养补充,维持氨基酸良好比值,对于运动员维持良好体能,延缓运动疲劳,促Objective To study the serum metabonomic changes of wrestlers before and after competition using the ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS)technique,so as to provide a new way for scientific nutrition supplement for rapid recoveryand identification of new training monitoring indexes.Methods Eleven female wrestlers(age,22.09±2.77 years;height,1.63±0.04 m;weight,60.45±8.43 kg;athlete levels,first-class athletes or elite athletes;mean±SD)were selected.They were collected blood samples from the cubital vein on an empty stomach at 7 a.m.one day before and after the competition.Two tubes of elbow venous blood were collected each time,one tube(3 mL)for whole blood biochemical indexes analysis,the other(5 mL)centrifuged for serum to undergo biochemical indexes detection and metabolomics testing.UPLC-Q-TOF-MS was used for metabolomics detection and data analysis to determine the metabolic differences before and after the competition.The obtained differential metabolites were analyzed to determine the potential changes in the metabolic pathway after the competition.Meanwhile,Pearson correlation analysis was conducted between biochemical indexes and metabolites.Results(1)The average serum urea(BU)and total testosterone(TT)levels were significantly higher after the competition(P<0.01,P<0.05),while the average hematocrit(HCT)was significantly lower than before the competition(P<0.05).(2)According to the criteria of P value less than 0.05 for comparison between groups and VIP(variable importance in projection)value greater than 1 for orthogonal partial least squares analysis,42 metabolic characteristic peaks were determined,and 19 differentially related metabolites were screened out,among which 13 metabolites were up-regulated and 6 down-regulated after the competition.(3)Two abnormal pathways after the competition were identified,namely,the phenylalanine metabolism pathway and the tryptophan-phenylalanine-tyrosine synthesis pathway.(4)Hemoglobin,HCT,BU,serum crea
