摘  要:从板蓝根多糖(Isatidis Radix polysaccharides, IRPS)对马立克病病毒(Marek?s disease virus, MDV)的增殖抑制、感染阻断以及直接杀灭这3个给药途径探讨IRPS抗MDV作用。利用Real-time quantitative PCR(RT-qPCR)、蛋白免疫印迹(Western blot)以及间接免疫荧光(IFA)等试验观察IRPS对病毒复制的影响。结果显示,IRPS剂量依赖性抑制MDV的基因表达、蛋白合成以及降低病毒复制水平。同时,IRPS可以降低IL-6的表达,并促进IFN-β的表达。结果表明,IRPS可能通过降低炎症反应,并且促进干扰素表达而抑制MDV的复制。To investigate the effect of Isatidis Radix polysaccharides(IRPS)on the replication of Marek's disease virus(MDV)in chicken embryonic fibroblast(CEF)cells.Here we tested the effect of IRPS on viral replication through pathways of proliferation inhibition,infection blockage and direct inactivation of MDV.Using Real-time quantitative PCR(RT-qPCR),Western blot,and indirect immunofluorescence assays,we found that IRPS has dose-dependence inhibitory effect on viral gene expression,protein synthesis and viral replication.Meanwhile,IRPS reduced expression of IL-6 and enhanced expression of IFN-β.These results indicate that IRPS may inhibit MDV replication through inhibiting inflammation response and increasing interferon response.Collectively,this study for the first time reveals inhibitory effect of IRPS on MDV replication in vitro,which provides research basis for MD prevention and control.
