摘 要:人工合成一种细胞穿膜肽,制备穿膜肽-DNA疫苗复合物,先将其转染Vero细胞,采用间接免疫荧光抗体试验(IFTA)鉴定目的基因在Vero细胞中的表达;再将40只6周龄雌性昆明小鼠分为CPPs-pVAX1-P33、pVAX1-P33、pVAX1和PBS 4个组;分别肌肉注射相应免疫原,采用ELISA方法检测CPPs血清中IgG抗体水平和IL-4、IFN-γ细胞因子水平;在三免2周后检测小鼠脾脏中CD3^(+)、CD4^(+)和CD8^(+)含量。结果显示,CPPs-pVAX1-P33复合物成功转染到Vero细胞,并在其中获得表达;CPPs-pVAX1-P33免疫组昆明小鼠血清中IgG抗体水平,IL-4和IFN-γ细胞因子水平及淋巴细胞亚群CD3^(+)、CD4^(+)、CD8^(+)含量均显著或极显著高于pVAX1-P33(P<0.05或P<0.01)。结果表明,本试验制备的CPPs能显著诱导瑟氏泰勒虫DNA疫苗的体液免疫和细胞免疫应答反应。The objective of this study was to evaluate the regulatory effect of a kind of cell-penetrating peptide on Thaleria serneri DNA vaccine.The cell transmembrane peptides(CPPs)were synthesized and the CPPs-DNA vacine complex was prepared.The CPPs-DNA vaccine complex was first transfected into Vero cells,and the expression of the target gene in Vero cells was identified by indirect immunofluorescence antibody test(IFAT).Forty 6-week-old female kunming mice were divided into four groups:CPPs-pVAX1-P33,pVAX1-P33,pVAX1 and PBS.The levels of IgG antibody and IL-4 and IFN-γcytokines in serum of CPPs were detected by ELISA.The levels of CD3^(+),CD4^(+)and CD8^(+)in spleen of mice were detected two weeks after the triple immunization.The results showed that CPPs-pVAX1-P33 complex was successfully transfected and expressed in Vero cells.The levels of IgG antibody,IL-4 and IFN-γcytokines and the contents of CD3^(+),CD4^(+)and CD8^(+)of lymphocyte subsets in CPPs-pVAX1-P33 group were significantly higher than those in pVAX1-P33 group(P<0.05 or P<0.01).The results showed that CPPs could significantly induce humoral and cellular immune responses of DNA vaccine.
