在LPS诱导的天然免疫反应中前列腺素D_(2)与TLR2和TLR4的相互调控作用

摘　　要：体外培养小鼠腹腔巨噬细胞后通过实时荧光定量PCR、Western blot、ELISA和流式细胞仪检测,分析模式识别受体TLR2和TLR4对脂多糖(lipopolysaccharides,LPS)诱导巨噬细胞前列腺素D合成酶表达和PGD_(2)分泌的影响。结果显示,在TLR2或TLR4缺失的情况下,LPS诱导巨噬细胞中H-PGDS和COX-2基因或蛋白的表达(P<0.05),以及PGD_(2)的分泌(P<0.01)会受到不同程度的影响。此外,分析了内外源性PGD_(2)对LPS诱导巨噬细胞TLR2和TLR4表达的影响。结果显示,前列腺素D合成酶抑制剂HQL-79和H-PGDS inhibitorⅠ预处理可显著下调LPS诱导TLR2的表达(P<0.001)。但是,当使用外源性PGD_(2)后,LPS诱导巨噬细胞TLR4的表达水平发生显著下调,而不是TLR2(P<0.05)。结果表明,TLR2和TLR4参与LPS诱导巨噬细胞中PGD_(2)合成分泌,且内外源性PGD_(2)对LPS刺激巨噬细胞诱导TLR2和TLR4的表达具有不同调控作用。The effects of TLR2 and TLR4 on the expression of prostaglandin D synthetase and PGD_(2)secretion in LPS-induced mouse peritoneal macrophages by RT-PCR,Western blot,ELISA and flow cytometry.The results showed that the impaired expression of H-PGDS and COX-2 genes or proteins and secretion of PGD_(2)in LPS-induced deficiency of TLR2 and TLR4 macrophages.In addition,the effects of exogenous and exogenous PGD_(2)on the expression of TLR2 and TLR4 in induced-LPS macrophages were analyzed.The results showed that pretreatment with prostaglandin D synthase inhibitor HQL-79 and H-PGDS inhibitor I significantly down-regulated the expression of TLR2 in LPS-induced macrophages(P<0.001).However,when exogenous PGD_(2)was used,the expression of TLR4 was significantly down-regulated,but not TLR2 in LPS-induced macrophages(P<0.05).These results suggest that TLR2 and TLR4 are involved in the synthesis and secretion of PGD_(2)in LPS-induced macrophages,and exogenous PGD_(2)has different regulatory effects on the expression of TLR2 and TLR4 in LPS-induced macrophages.
