摘  要:通过CCK-8法检测蚯蚓提取物和脂多糖(lipopolysaccharide, LPS)对RAW 264.7细胞活性的影响,用不同质量浓度的蚯蚓提取物预处理RAW 264.7细胞后用LPS刺激诱导炎症反应,利用实时荧光定量PCR(qPCR)检测TNF-α、IL-1β、IL-6和IL-10的转录水平,通过Griess法检测一氧化碳(NO)生成,Western blot检测诱导型一氧化氮合酶(iNOS)、p65、p-p65、iκBα、p-iκBα的蛋白表达。结果显示,当蚯蚓提取物≤4 mg/L时没有细胞毒性,1~4 mg/L蚯蚓提取物可减少炎症因子TNF-α、IL-1β、IL-6的产生,提升抑炎因子IL-10的分泌,显著抑制炎症介质NO及相关合成酶iNOS蛋白,以及抑制NF-κB的活化。结过表明,蚯蚓提取物具有显著的抗炎症作用,揭示蚯蚓提取物可用于治疗及预防部分炎症性疾病。The effects of earthworm extract and lipopolysaccharide(LPS)on the activity of RAW264.7 cells were detected by CCK-8 method.RAW264.7 cells were pretreated with earthworm extract of different concentrations,and then the inflammatory reaction was induced by LPS stimulation.TNF-α,IL-1β,IL-6 and IL-10 transcription levels were detected by quantitative real-time PCR(qPCR),Griess detection of carbon monoxide(NO)production,Western blot detection of inducible nitric oxide synthase(iNOS),p65,p-p65,IkBa,p-ikBαprotein expression were performed.There was no cytotoxicity when the concentration of earthworm extract≤4 mg/L,and 1-4 mg/L earthworm extract could reduce the production of the inflammatory factor TNF-α,IL-1β,IL-6,increase the secretion of anti-inflammatory factor IL-10,significantly inhibit inflammatory mediator NO and related synthase iNOS,and inhibit NF-B activation.It is proved that earthworm extract has significant anti-inflammatory effect,revealing that earthworm extract can be used to treat and prevent some inflammatory diseases.
