摘  要:利用原代奶牛肝细胞进行体外试验,首先用0.0,0.6,1.2,2.4 mmol/L非酯化脂肪酸(non-esterified fatty acids,NEFA)处理奶牛原代肝细胞,通过Western blot检测各组IL-11和凋亡关键蛋白cleaved Caspase-3的表达,qRT-PCR检测IL-11 mRNA表达。结果显示,在高NEFA刺激下,1.2,2.4 mmol/L NEFA可显著升高IL-11和cleaved Caspase-3蛋白的表达,同时显著上调IL-11 mRNA表达;然后,通过短发夹RNA(sh-RNA)沉默IL-11,并用1.2 mmol/L NEFA处理奶牛原代肝细胞,通过Western blot检测IL-11和cleaved Caspase-3蛋白表达,qRT-PCR检测IL-11 mRNA表达,结果显示沉默IL-11可显著抑制NEFA诱导的cleaved Caspase-3的表达升高。结果表明,IL-11可通过促进cleaved Caspase-3的表达介导高NEFA诱导的奶牛原代肝细胞凋亡。First,primary bovine hepatocytes were treated with 0.0,0.6,1.2,2.4 mmol/L NEFA,and the expression of IL-11 and the marker molecule of apoptosis-cleaved Caspase-3 was measured by Western blot.qRT-PCR was applied to measure the mRNA expression of IL-11.The results showed that upon high concentrations of NEFA stimulation,1.2,2.4 mmol/L NEFA could significantly increase the protein expression of IL-11 and cleaved Caspase-3,and significantly increase the mRNA expression of IL-11.Short hairpin RNA(sh-RNA)was used for silencing IL-11,and the cells were treated with 1.2 mmol/L NEFA.The protein expression of IL-11 and cleaved Caspase-3 were measured by Western blot,and mRNA expression of IL-11 was measured by qRT-PCR.Results showed that the increase of cleaved Caspase-3 protein expression induced by high NEFA was significantly attenuated by silencing IL-11 in bovine hepatocytes.Taken together,the results suggest that IL-11 could mediate high NEFA-induced hepatocyte apoptosis in dairy cows through promoting the expression of cleaved Caspase-3.
