摘  要:使用褪黑素处理犊牛睾丸支持细胞,观察细胞的状态,实时荧光定量RT-PCR和Western blot检测细胞增殖相关基因(PI3K、Akt)、细胞紧密连接相关基因(ZO-1、Connexin-34)mRNA和蛋白表达的变化,免疫荧光染色分析ZO-1的表达。结果显示,50μmol/L褪黑素显著促进支持细胞的增殖,PI3K、Akt的mRNA和蛋白表达水平显著升高,下游的相关基因CDK4、cyclinD1和cyclinE1的mRNA表达水平显著升高,P21Cip1的mRNA表达水平显著下降(P<0.05),ZO-1、Connexin-34的mRNA和蛋白表达水平没有显著变化(P>0.05),ZO-1的免疫荧光染色没有显著变化。结果表明,褪黑素通过PI3K/Akt信号通路促进犊牛睾丸支持细胞的体外增殖。To investigate the effects of melatonin on cell proliferation and its underlying mechanisms in sertoli cells,we treated the sertoli cells with melatonin and observed the state of the cells.Real-time fluorescent quantitative RT-PCR and Western blot were used to detect the mRNA and protein expression changes of cell proliferation related genes(PI3K and Akt)and cell tight junction related genes(ZO-1 and Connexin-34),and immunofluorescence staining was used to analyze the expression of ZO-1.The results showed that 50μmol/L melatonin significantly promoted cell proliferation in sertoli cells.Furthermore,melatonin significantly increased the mRNA and protein expression levels of PI3K and Akt,as well as the mRNA expression levels of downstream related genes CDK4,cyclinD1 and cyclinE1,and melatonin significantly(P<0.05)decreased the mRNA expression level of P21Cip1.There were no significant changes(P>0.05)in mRNA and protein expression levels of ZO-1 and Connexin-34,or in immunofluorescence staining of ZO-1.In summary,the results indicate that melatonin promotes the proliferation of sertoli cells in vitro through PI3K/Akt signaling pathway.
