摘  要:目的探讨紫花草总黄酮对大鼠尿路结石的改善及对尿路炎症损伤的影响。方法选取50只SD大鼠,将40只大鼠建立尿路结石模型,并按照体质量排序法分为模型组及紫花草总黄酮低、中、高剂量组,每组各9只;另取10只大鼠为对照组。检测各组大鼠的24h尿量、24h尿草酸排泄量、24h尿钙排泄量、尿酸、尿素氮、肌酐、白细胞介素-8(IL-8)、IL-1β、肿瘤坏死因子-α(TNF-α)、肾脏组织病理变化、钙盐沉积的情况;检测骨形态发生蛋白2(BMP2)、核心结合因子αl(Cbfα1)、锌指结构转录因子(Osx)蛋白的相对表达量。结果与对照组比较,模型组的24h尿量减少,24h尿草酸排泄量、24h尿钙排泄量增多,尿酸、尿素氮、肌酐、IL-8、IL-1β、TNF-α及BMP2、Cbfαl、Osx蛋白相对表达量升高(均P<0.05);与模型组比较,紫花草总黄酮低、中、高剂量组的24h尿量依次增多,24h尿草酸排泄量、24h尿钙排泄量依次减少,尿酸、尿素氮、肌酐、IL-8、IL-1β、TNF-α及BMP2、Cbfα1、Osx蛋白相对表达量依次降低(均P<0.05);与模型组比较,紫花草总黄酮低、中、高剂量组的肾小管上皮细胞水肿减轻,红染物质、炎性细胞浸润减少,钙盐沉积减少,其中高剂量组的改善更明显。结论紫花草总黄酮可减轻尿路结石大鼠的症状,改善肾功能,控制尿路炎症损伤,作用机制可能与抑制BMP2信号通路有关。Objective To investigate the effect of total flavonoids of arnebia on the improvement of urinary calculi and the injury of urinary tract inflammation in rats.Methods Ten of the 50 SD rats were randomly selected as the control group,and the other 36 urinary calculi model rats were randomly divided into model group and low,medium and high dose groups of total flavonoids of arnebia.24 h urine output,24 h urine oxalic acid excretion,24 h urine calcium excretion,uric acid,urea nitrogen,creatinine,interleukin IL-8 and IL-1β,Tumor necrosis factor-α(TNF-α)and renal histopathological changes and calcium salt deposition were measured.The relative expression of bone morphogenetic protein 2(BMP2),core binding factorα1(Cbfa1)and osterix(Osx)protein were measured.Results Compared with the control group,the 24 h urine volume in the model group decreased,the 24 h urine oxalic acid and calcium excretion increased,and the uric acid,urea nitrogen,creatinine,IL-8 and IL-1β,TNF-αand BMP2,Cbfαl,Osx protein increased(all P<0.05).Compared with the model group,the 24 h urine volume in the low,medium and high dose groups of total flavonoids of arnebia increased in turn,24 h urine oxalic acid and urinary calcium excretion decreased in turn,Uric acid,urea nitrogen,creatinine,IL-8,IL-1β,TNF-αand BMP2,Cbfal,Osx protein decreased in turn(all P<0.05).Compared with the model group,the edema of renal tubular epithelial cells,the infiltration of red dye and inflammatory cells,and the deposition of calcium salt were reduced in the low,medium and high-dose groups of total flavonoids of arnebia,especially in the high dose total flavonoids of arnebia group.Conclusions Total flavonoids of arnebia can reduce the symptoms of rats with urinary calculi,improve renal function and control urinary tract inflammatory injury.The mechanism may be related to the inhibition of BMP2 signal pathway.
