摘　　要：目的探究转录因子TCF-1对HIV-1特异性CD8^(+)T细胞免疫应答的调节作用。方法收集HIV-1感染者的外周血,使用HIV-1特异性抗原肽刺激病人PBMC(外周血单个核细胞),应用流式细胞术检测CD8^(+)T细胞的数目、TCF-1和T细胞耗竭相关的标志物的表达水平,定量PCR技术检测外周血的HIV-1病毒滴度。结果本研究发现,HIV-1慢性感染者的HIV-1特异性CD8^(+)T细胞中的TCF-1高表达的细胞亚群具有较好的细胞效应功能、较低的抑制性受体TIM3表达水平,并且TCF-1+CD8^(+)T细胞数目与血清中的HIV-1病毒载量呈负相关关系。结论TCF-1对病毒特异性CD8^(+)T细胞控制HIV-1感染具有重要的作用。This research is aimed at investigating the role of T cell factor-1(TCF-1)transcription factor in the immune response of HIV-1-specific CD8^(+)T cells.PBMCs from HIV-1 infected patients were collected and stimulated with HIV-1 antigen peptides,and then the expression of TCF-1 as well as T cell exhaustion and the number of CD8^(+)T cells were analyzed by flow cytometry.Furthermore,the HIV-1 virus titer in peripheral blood was detected by quantitative PCR.Data showed that the TCF-1 high-expression cell subsets in HIV-1-specific CD8^(+)T cells exhibited better cell effector function and lower expression levels of inhibitory receptor TIM3.Additionally,the number of TCF-1+CD8^(+)T cells was negatively correlated with the HIV-1 viral load in serum.In this study,our data demonstrated that TCF-1+HIV-1-specific CD8^(+)T cells play an important role in the control of HIV-1 infection.
