摘 要:目的探讨大孔树脂纯化丹参中丹酚酸B的工艺过程和吸附原理。方法根据正交试验设计优化丹酚酸B的提取工艺,采用大孔树脂纯化技术方法,考察上样流速、静态吸附时间、上样样品pH值及洗脱剂浓度、洗脱流速等因素对纯化效果的影响,同时分析大孔树脂对丹酚酸B的吸附动力学过程。结果丹酚酸B的最佳提取条件:70%乙醇为溶剂,提取2 h,提取1次。筛选D101型大孔树脂对丹酚酸B的最佳纯化条件:样品溶液pH值为4,吸附流速1 BV/h,静态吸附6 h,70%乙醇为洗脱剂,洗脱流速1 BV/h。通过动力学分析,准二级速率方程能更好描述D101型大孔吸附树脂对丹酚酸B的吸附过程,并且此过程受到薄膜扩散和内扩散的共同影响。结论优化得到的丹酚酸B提取纯化工艺条件可行有效,可为含有丹参的复方及丹酚酸B的制剂的优化开发研究提供参考。Objective:To explore and analyze the purification process and adsorption principle of salvianolic acid B from salvia miltiorrhiza by macroporous resin.Methods:According to the orthogonal experimental design,the extraction process of salvianolic acid B was optimized and macroporous resin was used to purify salvianolic acid B.The effects of loading flow rate,static adsorption time,p H of loading sample,eluent concentration and elution flow rate on the purification were taken into account to conduct investigation;simultaneously,the adsorption dynamics of salvianolic acid B by macroporous resin was analyzed.Results:The optimum extraction conditions of salvianolic acid B were as follows:70%ethanol as solvent,extracting for 2 h,extracting once.The optimum purification conditions of salvianolic acid B by D101 macroporous resin were as follows:pH of sample was 4,adsorption flow rate was 1 BV/h,static adsorption for 6 h,70% ethanol was used as eluent,the elution flow rate was 1 BV/h.The pseudo-second order kinetic equation could better describe the adsorption process of salvianolic acid B by D101macroporous resin,and this process was affected by both film diffusion and internal diffusion.Conclusion:The optimized extraction and purification conditions of salvianolic acid B are feasible and effective,which can provide a reference for the optimization and development of preparations of compound containing salvia miltiorrhiza and salvianolic acid B.
