摘 要:目的 探讨黏蛋白13(Mucin13,MUC13)对人喉鳞癌细胞增殖、化疗药物5-氟尿嘧啶细胞毒性、周期、凋亡、迁移、侵袭能力等生物学行为的影响。方法 将MUC13-RNAi序列包装入慢病毒并转染人喉鳞癌细胞株TU177,特异性阻低MUC13的表达。利用CCK-8法、平板克隆实验、Transwell小室迁移实验和流式细胞术观察MUC13对人喉鳞癌细胞TU177的增殖、化疗药物5-氟尿嘧啶细胞毒性、周期、凋亡、迁移、侵袭能力的影响。结果 与空白对照组(Blank组)及阴性对照组(NC组)相比,MUC13-RNAi组TU177细胞CCK-8测试光密度(OD)值增大率随培养时间延长而下降,化疗药物5-氟尿嘧啶对MUC13-RNAi组TU177细胞毒性增强;MUC13-RNAi组较Blank组TU177细胞相对克隆形成率为54.1%(P<0.05);迁移实验中,MUC13-RNAi组与Blank组相比TU177细胞相对迁移率为53.2%,MUC13-RNAi组与NC组相比TU177细胞相对迁移率为56.3%,差异均有统计学意义(P值均<0.05);侵袭实验中,MUC13-RNAi组与Blank组相比喉癌细胞相对迁移率为65.9%,MUC13-RNAi组与Blank组相比喉癌细胞相对迁移率为69.2%,差异均有统计学意义(P值均<0.05);同时MUC13-RNAi组TU177细胞发生G1期阻滞及早期凋亡倾向增加,表明MUC13-RNAi组TU177细胞增殖能力下降。结论 特异性阻低MUC13在喉鳞癌细胞系TU177中的表达后,细胞的增殖能力下降、化疗药物5-氟尿嘧啶对细胞毒性增强、细胞迁移能力下降、细胞稍有G1阻滞并且凋亡倾向增加,这表明MUC13参与调控了喉鳞癌细胞的多种生物学行为,具有作为喉鳞癌治疗靶标的潜力。Objective To determine the affection of Mucin 13(MUC13)on the biological behaviors such as proliferation,drug sensitivity,cell cycles,apoptosis,migration,and invasion of cells of human laryngeal squamous cell carcinoma.Methods MUC13-RNAi sequence was packaged into lentivirus and transfected into human laryngeal squamous cell carcinoma cell line TU177.CCK-8 assay,tablet cloning assay,transwell assay,and flow cytometry were applied to confine the affection of MUC13 expression inhibition on the proliferation,drug sensitivity,cycle,migration,invasion behaviors of TU177 cells.Results Compared with the Blank control(Blank)TU177 cells and the meaningless sequence transfected negative control(NC),the OD value variation rate of CCK-8 test of TU177 cells in the MUC13-RNAi group decreased with culture time,cytotoxicity of 5-Fluorouracil to TU177 cells increased.The relative clone formation rate of Tu177 cells in the NC group was 78.9%and that of TU177 cells in the MUC13-RNAi group was 54.1%compared with that in the blank group(P<0.05).In the migration experiment,the relative migration rate of TU177 cells in the MUC13-RNAi group was 56.3%when compared with that of NC group,with statistical significance(P<0.05).In the invasion experiment,the relative migration rate of laryngeal cancer cells in the MUC13-RNAi group was 69.2%when compared with that of NC group,the differences were statistically significant(P<0.05).At the same time,TU177 cells in the MUC13-RNAi group experienced G1 phase arrest and increased tendency of early apoptosis.Conclusions After the specific inhibition of MUC13 expression in the laryngeal squamous cell carcinoma line TU177,the proliferation ability,cytotoxicity of 5-Fluorouracil to TU177 cells,and migration ability of the cells were decreased,the cells were slightly G1 blocked and the apoptotic tendency was increased,suggesting that MUC13 is involved in the regulation of a variety of biological behaviors of laryngeal squamous cell carcinoma cells.
