摘  要:探讨不同pH值和盐条件下混合模式层析介质(MMA Focurose 6FF)与离子交换层析介质(DEAE Fo‐curose 6FF)的牛血清白蛋白(BSA)吸附性能.结果显示:1)未加盐时,两种介质均在BSA等电点附近有最大饱和吸附容量Q_(m),当pH值偏离等电点,Q_(m)下降,表观解离常数Kd升高;2)添加NaCl后,MMA介质的BSA吸附受盐浓度影响较小,中低盐条件下有较高Q_(m),DEAE介质在低盐时可吸附BSA,但随NaCl浓度升高,Q_(m)急剧下降,高盐下几乎不吸附;3)添加(NH_(4))_(2)SO_(4)后,MMA介质出现“U”型吸附规律,0.50~0.75 mol/L时Q_(m)最低,高盐时Q_(m)可恢复60%以上,且pH值越高,恢复比例越大,DEAE介质基本不吸附BSA.结果表明,混合模式介质具更佳的耐盐吸附性能,本结果可为蛋白分离设计提供参考.This paper discusses the adsorption performances of bovine serum albumin(BSA)on mixed-mode chromatography resin(MMA Focurose 6FF)and ion exchange chromatography resin(DEAE Focurose 6FF)under different pH value and salt conditions.The results show that:1)When no salt is added,the maximum saturated adsorption capacities(Q_(m))are found near the isoelectric point of BSA for the two resins.When the pH value deviates from the isoelectric point,Q_(m)decreases and the apparent dissociation constant(Kd)increases.2)When NaCl is added,the BSA adsorption on MMA resin is less affected by the salt addition.Moreover,higher Q_(m)values are found under the medium and low salt concentrations.DEAE resin can adsorb BSA at low salt concentration,but Q_(m)decreases sharply with the increase of NaCl concentration.In addition,it can hardly adsorb BSA at high salt concentration.3)When(NH_(4))_(2)SO_(4)is added,MMA resin shows a"U"type adsorption trend.The lowest level of Q_(m)appears in the range of 0.50~0.75 mol/L(NH_(4))_(2)SO_(4).Besides,Q_(m)recovers more than 60%at high salt condition.The higher the pH is,the greater the recovery ratio is.Interestingly,DEAE resin can hardly adsorb BSA at the same conditions.The results indicate that mixed-mode resin have higher salt tolerance,which is expected to provide reference for the design of protein separation.
