摘 要:目的 优化NabTM技术制备柚皮素的牛血清白蛋白纳米粒的处方工艺。方法 以包封率为指标,采用Plackett-Burman实验设计结合Box-Behnken响应面法优化柚皮素白蛋白纳米粒的处方工艺,并考察纳米粒的体外释放规律。结果 最终确定柚皮素白蛋白纳米粒处方工艺为:白蛋白浓度为30 mg·mL-1,水相pH值为6,有机相与水相比例为1∶5,制备的纳米粒包封率为88.80%,粒径为171.9 nm, PDI为0.340,体外释放具有明显缓释作用。结论 Plackett-Burman实验设计联用Box-Behnken响应面法可用于优化柚皮素白蛋白纳米粒工艺处方。OBJECTIVE To optimize the formulation of bovine serum albumin nanoparticles(BSA)for naringenin preparation by Nab TM technology.METHODS The formulation and technology of naringenin albumin nanoparticles were optimized by Plackett Burman experimental design combined with Box Behnken response surface methodology,and the in vitro release of naringenin albumin nanoparticles was investigated.RESULTS The formulation of naringenin albumin nanoparticles was:the concentration of albumin was 30 mg·mL-1,the pH of aqueous phase was 6,and the ratio of organic phase to aqueous phase was 1∶5.The entrapment efficiency of the prepared nanoparticles was 88.80%,the particle size was 171.9 nm,and the PDI was 0.340.The in vitro release of the nanoparticles showed significant sustained release effect.CONCLUSION Plackett Burman experimental design combined with Box Behnken response surface methodology can be used to optimize the formulation of naringenin albumin nanoparticles.
