摘 要:为研究羊驼(Vicugna pacos)皮肤中控制黑色素生成的鼠灰色基因(Agouti Signaling gene,Agouti)和前黑素小体蛋白基因(premelanosome protein gene,PMEL)的表达量与毛纤维中黑色素含量的相关性,试验从不同毛色羊驼皮肤差异表达基因中筛选主要毛色相关基因。用紫外分光光度法测定白色、棕色和黑色羊驼毛纤维中的黑色素含量,并用实时定量PCR(qRT-PCR)法和Fisher法对Agouti与PMEL的相对表达量与毛纤维中黑色素的含量的相关性进行分析。结果显示,23个毛色基因在白色和黑色羊驼皮肤中呈差异表达,白色和褐色皮肤中Agouti的mRNA表达显著高于黑色皮肤中Agouti的表达;PMEL在羊驼黑色皮肤中的表达高于棕色皮肤和白色皮肤中的表达(P<0.05)。Agouti与PMEL的表达量均与毛纤维中黑色素含量相关。本研究为揭示羊驼毛色的分子机制提供理论基础,为分子育种提供了重要的理论依据。To investigate the correlation between the expression of Agouti Signaling gene(Agouti)and the premelanosome protein gene(PMEL)which control melanin production in the skin of alpaca(Vicuga pacos)and the melanin content in the hair with different colors,the transcriptome expression profiles of hair color related genes in alpaca skins with different hair colors were constructed.The contents of melanin in white,brown and black alpaca hair fibers were determined by ultraviolet spectrophotometry,and the correlation between the relative expression of Agouti and PMEL and the contents of melanin in hair fibers was analyzed by quantitative real-time PCR(qRTR-PCR)method and Fisher method.The results showed that 23 hair color related genes were differentially expressed in alpaca skins with white and black hair.The mRNA expression of Agouti was higher in skins with white and brown hair,but lower in black hair.The expression of PMEL in alpaca skins with black hair color was higher than that in skins with brown and white hair colors(P<0.05).The expression of Agouti and PMEL was related to the content of melanin.This study provides a theoretical basis for revealing the molecular mechanism of alpaca hair colors,and an important theoretical basis for molecular breeding.
