摘 要:目的:应用Box-Benhnken响应曲面设计优化超临界CO_(2)萃取人参叶中人参叶总皂苷的工艺条件。方法:利用超临界CO_(2)技术提取人参叶中的总皂苷,采用紫外分光光度法对萃取物中总皂苷的含量进行测定,通过Box-Benhnken响应曲面设计进行多元线性回归和二项式拟合考察超临界CO_(2)萃取条件对人参总皂苷提取率的影响。结果:分光光度法测定波长为205 nm时,总皂苷质量浓度为0.0508~0.1016 mg/mL呈现良好的线性关系(r=0.9977),超临界萃取条件萃取温度为40℃,萃取压力为20 MPa,物料粒度为80目,乙醇夹带剂含量为0.25 mL时,人参叶中的人参总皂苷平均提取率为15.89%,平行测定三次最高提取率为16.07%,相对标准偏差为1.53%。结论:采用超临界CO_(2)萃取人参叶中人参总皂苷,高效、环保、工艺稳定,适用于工业化推广。Objective:To optimize the technological conditions of supercritical CO_(2) extraction of total saponins from ginseng leaves by box benhnken response surface design.Methods:Supercritical CO_(2) technology was used to extract total saponins from ginseng leaves,and the content of total saponins in the extraction was determined by UV spectrophotometry.Box-benhnken response surface design was used for multiple linear regression and binomial fitting to investigate the effect of supercritical extraction conditions on the extraction rate of total saponins of ginseng.Results:When the wavelength of spectrophotometry was 205 nm,the results showed that there was a good linear relationship in the range of the concentration of total saponins was 0.0508~0.1016 mg/mL(r=0.9977).The supercritical extraction conditions were as follows:extraction temperature 40℃,extraction pressure 20 MPa,particle size 80 mesh,ethanol entrainer 0.25 mL,the highest extraction rate of total saponins in ginseng leaves was 15.89%,the highest extraction rate was 16.07%after three parallel determinations,and the relative standard deviation was 1.53%.Conclusion:The supercritical CO_(2) extraction of total ginsenosides from ginseng leaves is efficient,environmental friendly and stable,which is suitable for industrial promotion.
