摘  要:目的:建立液相色谱-质谱联用仪测定洋葱中槲皮素的方法。方法:样品经过95%乙醇超声提取,BEH C_(18)色谱柱(2.5μm×100 mm,1.7μm),以0.1%甲酸-甲醇(35∶65)为流动相,流速为0.25 mL·min^(-1),柱温30℃,进样量1μL;ESI离子源,正离子模式,多反应监测模式,外标法定量。结果:槲皮素在10~250 ng·mL^(-1)线性关系良好,线性关系R^(2)=0.999。该方法检出限为0.01 mg·kg^(-1),定量限为0.05 mg·kg^(-1),加标回收率为92.0%~102.3%,精密度0.8%~3.1%。结论:本文所建立的方法操作简便、样品用量少、检出限低、准确度高,适用于洋葱中槲皮素的测定。Objective:To establish a liquid chromatography-mass spectrometry method for the determination of quercetin in onions.Method:The sample was extracted by ultrasound with 95%ethanol and subjected to BEH C_(18)(2.5μm×100 mm,1.7μm),with 0.1%formic acid methanol(35∶65)as mobile phase,flow rate of 0.25 mL·min^(-1),column temperature of 30℃,and injection volume of 1μL;ESI ion source,positive ion mode,multi reaction monitoring mode,external standard method for quantification.Result:Quercetin showed a good linear relationship between 10 ng·mL^(-1)and 250 ng·mL^(-1),with a linear relationship of R^(2)=0.999.The detection limit of this method is 0.01 mg·kg^(-1),the quantitative limit is 0.05 mg·kg^(-1),the recovery rate of spiking is 92.0%~102.3%,and the precision is 0.8%~3.1%.Conclusion:The method established in this article is simple to operate,with a small sample amount,low detection limit,and high accuracy,and is suitable for the determination of quercetin in onions.
