摘 要:采用甲醇超声和加热回流两种方式制得生地黄、熟地黄、生地炭和熟地炭等4种怀地黄炮制品,利用高效液相色谱法测定其梓醇、地黄苷A、地黄苷D、毛蕊花糖苷和5-羟甲基糠醛等有效成分的含量。生地黄经炮制后,其炮制品中的梓醇、地黄苷A、地黄苷D和毛蕊花糖苷含量减少,而5-羟甲基糠醛含量增加。甲醇超声制得的地黄炮制品中的梓醇、地黄苷D和毛蕊花糖苷含量高于加热回流法制得的地黄炮制品,而5-羟甲基糠醛含量低于加热回流法制得的地黄炮制品。超声处理生地黄中梓醇含量最高,为1.18%(超声提取),加热回流处理熟地炭中梓醇含量最低,为0.31%。甲醇超声制得的生地黄炮制品中地黄苷A、地黄苷D和毛蕊花糖苷含量分别为0.17%、0.65%和0.03%,而加热回流制得的熟地炭中5-羟甲基糠醛含量为0.23%。建议采用超声提取法测定地黄药材有效成分含量。Four processed products of Radix rehmanniae i.e.Raw Radix rehmanniae,Prepared Radix rehmanniae,biogenic charcoal and mature charcoal were prepared by methanol ultrasound and heating reflux.The contents of catalpol,Radix rehmanniae glycoside A,Radix rehmanniae glycoside D,mullein glycoside and 5-hydroxymethyl furfural were determined by HPLC.After processed,the contents of catalpol,Rehmannia glycoside A and D,and mullein glycoside in processed Rehmannia glutinosa decreased,while the content of 5-hydroxymethyl furfural increased.The contents of catalpol,Rehmannia glycoside D and mullein glycoside in processed Rehmannia glutinosa prepared by methanol ultrasonic method were higher than those prepared by heating reflux method,while the content of 5-hydroxymethyl furfural was lower than that of processed Rehmannia glutinosa prepared by heating reflux method.The highest catalpol content in raw Rehmannia glutinosa was 1.18%(ultrasonic extraction),and the lowest catalpol content in mature rehmannia charcoal was 0.31%(heating reflux extraction).The contents of Rehmannia glycoside A,D and mullein glycoside in raw Rehmannia processed by methanol ultrasound were 0.17%,0.65%and 0.03%respectively,while the content of 5-hydroxymethyl furfural in cooked Rehmannia charcoal prepared by heating reflux was 0.23%.It is suggested to use ultrasonic extraction method to determine the active ingredients content of Rehmannia glutinosa.
